    Notes: Summary Male albino rats treated with excessive amounts of pyridoxine developed an impairment of neuromuscular function. The equatorial region of the plantar lumbrical intrafusal muscle fibres was studied in the electron microscope and the calibre of the nerve fibres was determined in semi-thin sections of the posterior tibial nerves. Degeneration of the primary sensory endings coincided with the onset of ataxia, and in more advanced stages of the neuropathy as well as after a 2-month treatment-free period the equatorial region was denervated. There was a corresponding decrease in the number of large nerve fibres. It is considered essential that primary sensory endings of lumbrical muscle spindles should be included in studies of distally accentuated sensory neuropathies.

    Notes: Summary Neuronal responses to moving visual stimuli were recorded in the lateral geniculate nucleus (LGN) and area 17 of cats. Response duration (DE), number of spikes (NT), and mean frequency (FM) were estimated from the response histograms and analysed for their dependence on stimulus velocity. In the LGN, for about 2/3 of cells these response parameters changed monotonically with velocity up to about 100 °/s. In 1/3 of the cells, the response frequency was tuned to velocity. The speed at which individual cells reached a peak or plateau firing rate was correlated with their receptive field size. In area 17, most neurones were tuned to velocity. Nine out of 59 cells were insensitive to stimulus speed in that they responded equally well at stimulus velocities up to about 10 °/s. The results suggest that at higher levels in the nervous system information about velocity is represented in discrete groups of neurones. It is pointed out that different response parameters may be relevant for different perceptual phenomena associated with movement. The significance of integrational properties and lateral inhibition of nerve cells for the development of complex response properties is discussed.

    Notes: Summary Changes of specific response properties of single neurones in area 17 of the cat's cortex were studied during electrophoretic administration of glutamate and gamma-aminobutyric acid (GABA). Most cells were excited by glutamate. The diameter of the cells' discharge field (ERF) was enlarged by less than 1° in most cells, and the amplitude of the response within the ERF was increased. There was a weak correlation between the original size of the ERF and its enlargement during glutamate. Stimuli presented in non-optimal orientations and moved in the non-optimal direction became also effective in eliciting an excitatory response. When glutamate “doses” sufficiently high to increase background activity were used, inhibitory regions of the receptive fields could be revealed. GABA decreased the excitability of the cells until the response was totally blocked. In some cases, glutamate also decreased the excitability of cells. Simultaneous recordings from two cells suggested that the inhibitory effect may be indirect through activation of nearby inhibitory neurones. It is pointed out that extracellular recording combined with electrophoretic administration of glutamate can reveal information about a cell's properties which can otherwise only be obtained with intracellular recordings. The functional implications of the results are discussed.

    Notes: Abstract Cortisol and growth hormone (GH) secretion (spontaneous variations at night and the release induced by insulin hypoglycaemia) were investigated in 69 children and adolescents. Statistical analysis of approximately 600 pairs of cortisol and GH values in this study demonstrated that physiological fluctuations of cortisol do not alter GH secretion. A review of the literature shows that GH secretion is consistently depressed in Cushing's disease of central origin and in Cushing's syndrome due to adrenal carcinoma. When acutely administered, doses higher than 100 mg of cortisol (or equivalent amounts of other steroids) per adult are necessary to block GH secretion and the hormones have to be given several hours previously. In long-term steroid treatment, suppression of GH is observed in only 1 out of 3 patients. The effect apparently does not persist beyond elimination of the last dose, i.e. generally not longer than 12 to 24 h. These data can be taken as a rationale for intermittent or alternating dosage schedules, and for the use of short acting derivatives if long-term, high-dose steroid treatment is necessary in children. It remains to be established whether growth deficiency in exogenous hypercortisolism is due to suppression of GH secretion, decreased production of somatomedins, direct antagonism of the action of somatomedins on growing cartilage, or a combination of these mechanisms.

Dagegen lassen sich kürzer dauernde Bestimmungen des Blutvolums mit dem Morawitzschen Verfahren auch hier anstellen und ergeben ähnliche Werte wie beim Erwachsenen. 3. Mit der gleichen Technik ist es möglich, die Volumänderung der Gefäße bei verschiedener Temperatur des umgebenden Wassers zu untersuchen. 4. In einem Fall von Morbus Raynaud ließen sich die Befunde Curschmanns bestätigen: Die normale Gefäßreaktion zeigte Störungen. Dagegen fand sich keine Abweichung des direkten Temperatureinflusses auf die Weite der Gefäße.

    Notes: Summary Plasma levels and urinary excretion of3H-fentanyl were studied in 5 human subjects after intravenous injection of this drug. After an initial rapid decline, the plasma level of fentanyl decreased slowly and approximately exponentially. The plasma concentration of metabolites remained almost steady from 1–3 h after injection. More than 60% of the administered radioactivity was excreted through the kidneys within 4 days. Only a small proportion of it was unchanged fentanyl. The rates of fall of plasma concentration and of urinary excretion were slower in man than in rabbits. — The time courses of plasma concentrations and of urinary excretion were simulated on an analogue computer. The results support the assumption that the different time courses of concentrations in man and rabbits are caused by slower metabolism in man. It seems likely that redistribution plays a dominant part in the short duration of action of fentanyl in man.

    Notes: Summary A simple method is described for the separate determination in plasma of lipophilic drugs and their more hydrophilic metabolites by liquid scintillation counting. This is done by measuring the radioactivity after adding biological specimens to a scintillation liquid which takes up both the unchanged drug and its hydrophilic metabolites (Bray's solution), and by mixing further aliquots of the specimen with a scintillation fluid which only takes up unchanged drug (Toluene+PPO=TSC). The amount of hydrophilic substances dissolved in TSC, as well as irradiation from the aqueous phase, were shown to be less than 13% for both of the isotopes3H and14C, employing 0.5 ml of aqueous phase added to 10 ml toluene scintillant. Checking the method by thin layer chromatography showed that the concentrations of fentanyl and haloperidol in plasma were determined accurately. After taking account of a special procedure used to dissolve a greater proportion of morphine in toluene, the ratio of TSC and BSC measurements was in agreement with data obtained by thin layer chromatography. Experiments with chlorpromazine showed that lipophilic degradation products were formed which could not be separated from the original drug by liquid-liquid partition. In such cases, therefore, the method cannot be employed. Etorphine is an example of how reliable results were obtained despite low dosage and relatively low specific activity of this drug.
