    Notes: Abstract Clinical data and autopsy findings in a case ¶of new variant Creutzfeldt-Jakob disease (vCJD) are reported. this case, the first histologically confirmed case described outside the United Kingdom, very much resembles the cases described by Will et al. [(1996) Lancet 347 : 921–925] and Zeidler et al. [(1997) Lancet 350 : 903–908, 908–910]. Neuropathological studies failed to reveal any conspicuous clues that could be relevant for understanding the pathophysiology of the disease. For epidemiological surveillance, neuropathologists should scrutinize suspected cases keeping in mind the possibility of vCJD.

    Notes: Abstract In a greenhouse pot study, we examined the availability of N to grain sorghum from organic and inorganic N sources. The treatments were15N-labeled clover residues, wheat residues, and fertilizer placed on a sandy clay loam and loamy sand soil surface for an 8-week period. Soil aggregates formed under each soil texture were measured after 8 weeks for each treatment. Significantly greater 15N was taken up and recovered by grain sorghum in sandy clay loam pots compared with loamy sand pots. Greater 15N recovery was consistently observed with the inorganic source than the organic sources regardless of soil texture or time. Microbial biomass C and N were significantly greater for sandy clay loam soil compared with the loamy sand. Microbial biomass 15N was also significantly greater in the sandy clay loam treatment compared to the loamy sand. The fertilizer treatment initially had the greatest pool of microbial biomass 15N but decreased with time. The crop residue treatments generally had less microbial biomass 15N with time. The crop residues and soil texture had a significant effect on the water-stable aggregates formed after 8 weeks of treatments. Significantly greater water-stable aggregates were formed in the sandy clay loam than the loamy sand. Approximately 20% greater water-stable aggregates were formed under the crop residue treatments compared to the fertilizer only treatment. Soil texture seemed to be one of the most important factors affecting the availability of N from organic or inorganic N sources in these soils.

    Notes: Abstract In agricultural ecosystems that have had consistent cropping histories, standard microbial methods may be used to evaluate past and present practices. Our objective was to evaluate several microbial methods that best indicate cropping histories and soil quality on long-term plots. We selected soil microbial carbon (C), phospholipid analyses, direct counts of total fungal and bacterial biomass, and soil enzymes (phosphatases) to measure direct and indirect microbial activity on the Sanborn Field and Tucker Prairie. The Sanborn Field has been under various cropping and management practices since 1888 and the Tucker Prairie is an uncultivated site. Seven different plots were chosen on the Sanborn Field and random samples were taken in the summit area on the Tucker Prairie, which represented a reference site. Soil microbial biomass C, phospholipids, and enzyme activity were reflective of the cropping and management histories observed on the Sanborn Field. Enzymatic activity was highly correlated to soil organic matter. The direct counts of fungal and bacterial biomass showed that fungal populations dominated these soils, which may be attributed to soil pH. Soil microbial biomass C and enzyme assays seemed to be better potential indicators of cropping histories than the other methods tested in the long-term plots.

    Notes: Abstract  Earthworms may alter the physical, chemical, and biological properties of a forest soil ecosystem. Any physical manipulation of the soil ecosystem may, in turn, affect the activities and ecology of earthworms. The effects of removing organic matter (logs and forest litter) and severely compacting the soil on native earthworm species were measured in a central USA hardwood region (oak-hickory) forest in the Missouri Ozarks (USA). Soils in this region are characterized by a cherty residuum that is primarily of the Clarksville series (Loamy-skeletal, mixed, mesic Typic Paledults). Earthworms were collected from 0–15 cm depth each spring and fall for 2 years by handsorting, and densities were determined on a per meter square basis. Two native earthworm species, Diplocardia ornata and Diplocardia smithii, were dominant on this site. Organic matter removal decreased the average individual biomass of both species. However, both species responded differently to soil compaction. Soil compaction affected D. ornata adversely and D. smithii favorably. This suggested that the degree of soil compaction was not as restrictive with respect to D. smithii (2 mm diameter) as to D. ornata (5 mm diameter). Moreover, the apparently improved soil environmental conditions resulting from the remaining organic matter in compacted soil enhanced the population and growth of D. smithii. Sampling position on the landscape affected D. ornata but not D. smithii. Soil microbial biomass C and soil microbial biomass N were decreased under soil compaction when the organic matter was removed. Other factors influencing the ecology and activity of these two species will require further study.

    Notes: Abstract The interaction of vesicular-arbuscular mycorrhizae (VAM) and phosphate-solubilizing bacteria (PSB) on plant growth, soil microbial activities, and the production of organic acids was studied in non-sterile soil containing hydroxyapatite and glucose. Glomus etunicatum (VAM), a fungus, and Enterobacter agglomerans, a bacterium able to solubilize insoluble phosphate, were used as inocula. Three treatments and a control were used: inoculation with E. agglomerans (treatment E), inoculation with G. etunicatum (treatment G), inoculation with E. agglomerans+G. etunicatum (treatment E+G) and the control (C). Inoculation with E, G, or E+G had increased plant growth by days 35, 55, and 75 compared with the control. Microbial biomass carbon (C) and alkaline phosphatase activity in the rhizosphere generally increased with time. Alkaline phosphatase activity was higher in treatments G and E+G compared with the control at 35 and 55 days. The highest acid phosphatase activity was observed in treatment E at 35 days; however, this markedly decreased with time. A significantly higher soluble phosphorus (P) concentration was observed in treatments E and E+G on day 55 compared with C. However, there was no significant difference in soluble P concentration in the rhizosphere between treatments with time. The P concentration was greatest in all treatments on day 55. The highest oxalic acid concentration was observed in the rhizosphere of the non-sterile soil in E+G on day 35. Total N and P uptake in plants from treatments E and G were higher compared with the control. However, the highest N and P uptake was observed in treatment E+G. This study suggests a synergistic interaction between E. agglomerans and G. etunicatum.

    Notes: Abstract Phosphate-solubilizing bacteria (PSB) possessing the ability to solubilize insoluble inorganic phosphate were isolated from the rhizosphere soil of wheat. A laboratory study was conducted to investigate the solubilization of phosphate by a known PSB, Enterobacter agglomerans, and by a genetically manipulated bacterium, Escherichia coli. A second laboratory study investigated the release of P from E. agglomerans compared with known acids. For the first laboratory study, a cosmid (pHC79) library of phosphate-solubilizing gene(s) from E. agglomerans chromosome DNA was constructed in E. coli JM109. The clone JM109 (pKKY) showing phosphate solubilization properties was screened on standard medium containing hydroxyapatite (HY). The P concentration significantly increased at 5 and 10 days for JM109 (pKKY) compared with JM109 (pHC79), the control. Although the P concentration increased, there was no significant change in their pHs. Furthermore, an increase in colony-forming units (CFUs) was seen at 5 and 10 days for JM109 (pKKY) but not for JM109 (pHC79). Artificial acidification of the culture medium with HCl, citric acid, oxalic acid, and lactic acid was achieved by shaking for 48h. Acidification with these selected acids solubilized more HY than E. agglomerans growing for 42h at similar pHs. However, a high P concentration was measured in culture medium with E. agglomerans growing for 84h despite similar pHs. Our results suggest that acid production may play an important role in HY solubilization, but is not the sole reason for the increase in P concentration in culture medium.

    Notes: Summary The greater hip fracture rate among elderly women is generally ascribed to differences in femoral neck strength between the sexes. Strength of a given bone is a function of both its material properties and the magnitudes of mechanical stresses within it. This study examined the hypothesis that these apparent strength differences between the sexes are due to dissimilarities in the restructuring of the femoral neck with age, which result in higher stresses in elderly women. Using Hip Strength Analysis, a computer program developed by the authors, femoral neck cross-sectional geometric properties for stress analyses were derived from bone mineral image data of 409 community living, white subjects ranging from 19 to 93 years of age. Though both sexes show declines in femoral neck bone mineral density (BMD) and cross-sectional area with age, only females show a decline in the cross-sectional moment of inertia (CSMI, a geometric index of bone rigidity). The lack of decline in male CSMI appears to be a result of a small but significant increase in femoral neck girth. Similar age-related changes have been observed in the femoral shaft by others. The net effect of these observed changes is that mechanical stresses in the femoral neck of females appear to increase at three times the rate per decade of those of males. These results lend support to the hypothesis that the higher fracture rate in elderly women is due, at least in part, to elevated levels of mechanical stress, resulting from a combination of greater bone loss and less compensatory geometric restructuring with age.

A mutant strain of Salmonella thompson (S. thompson/r), with an increased resistance to gamma radiation, did not appear to possess a DNA repair system, active in a “non-growth” medium, similar to that reported for Micrococcus radiodurans and Escherichia coli B/r. 2. DNA strand breaks occurred to a similar degree in both the sensitive parent strain and the resistant mutant strain during exposure to gamma radiation. 3. The DNA could be reconstituted in both strains, but only when the cells were incubated in a growth-promoting medium after irradiation. 4. Results indicated that inactivation of S. thompson by gamma radiation was not the result of the production of DNA single-strand breaks per se, and the resistance of the mutant strain, therefore, did not appear to be directly related to the ability to repair such breaks.
