    Notes: Summary Although the involvement of taurine in osmoregulation is well-documented and widely accepted, no detailed mechanism for this function has been reported so far. We used subtractive hybridization to study mRNA steady state levels of genes up- or downregulated by taurine. Rats were fed taurine 100mg/kg body weight per day for a period of three days and hearts (total ventricular tissue) of experimental animals and controls were pooled and used for mRNA extraction. mRNAs from two groups were used for subtractive hybridization. Clones of the subtractive library were sequenced and the obtained sequences were identified by gen bank assignment. Two clones were found to contain sequences which could be assigned to the osmolarity sensor protein envZ, showing homologies of 61 and 65%. EnvZ is an inner membrane protein in bacteria, important for osmosensing and required for porine gene regulation. It undergoes autophosphorylation and subsequently phosphorylates OmpR, which in turn binds to the porin (outer membrane protein) promoters to regulate the expression of OmpF and OmpC, major outer membrane porines. This is the first report of an osmosensing mechanism in the mammalian system, which was described in bacteria only. Furthermore, we are assigning a tentative role for taurine in the osmoregulatory process by modifying the expression of the osmoregulatory sensor protein ENVZ.

    Description / Table of Contents: Zusammenfassung Der Einfluß einer Ganzkörper-Röntgenbestrahlung mit 440 rad auf die Aktivität des Enzyms Aldose-Reduktase (E.C. 1.1.1.21) in den Linsen von jungen Ratten wurde untersucht. Unmittelbar nach der Bestrahlung besteht kein Unterschied der Aktivität des Enzyms zwischen bestrahlten und unbestrahlten Linsen. Innerhalb von 10 Tagen entwickeln sich jedoch signifikante Unterschiede. Während die Aktivität in den Linsen der Kontrolltiere ständig ansteigt, bleibt sie in den bestrahlten Linsen nahezu konstant. Am 10. Tag nach der Bestrahlung zeigt das Enzym in beiden Versuchstiergruppen identische enzymkinetische Parameter. Es ist deshalb anzunehmen, daß die Bestrahlung das im Zeitpunkt der Strahleneinwirkung vorhandene Enzym nicht schädigt, seine Synthese dadurch aber quantitativ vermindert wird.
    Notes: Abstract The influence of whole body X-irradiation on the activity of the enzyme aldose reductase (E.C. 1.1.1.21) in the lenses of young rats was investigated. Immediately after irradiation there was no difference between the activity of the enzyme in irradiated and nonirradiated animals. Within 10 days after irradiation, significant differences developed. There was a constant increase of the activity in the control lenses, while the activity in the irradiated lenses remained nearly constant. There was no remarkable difference between the two groups 10 days after irradiation with respect to the kinetics of the enzyme. This meant that irradiation does not affect the enzyme present at the moment of application, but the influence of 440 rad X-irradiation effects an obvious decrease in the synthesis of the enzyme.

    Notes: Zusammenfassung Der Lungenkollaps ist solange ein physiologischer Zustand, wie die Verkleinerung des Lungenvolumens durch die elastischen Elemente der Lungenstruktur entstanden ist, die regulativen mechanismen voll wirksam bleiben, so daß die Zirkulation der Ventilation angepaßt werden kann und die Leistung des rechten Herzens weder in Ruhe noch nach Belastung unter Drucksteigerung zustande kommt. Pathophysiologisch wird der Lungenkollaps neben anderem in erster Linie durch Ausfall von Atemmuskulatur und durch Pleuraverschwartung, weil jetzt die Kollapslunge ungleichmäßig ventiliert und daher auch ungleichmäßig durchblutet wird. Das Angiogramm zeigt in Gebieten unter starrem Kollaps nicht nur eine Drosselung, sondern auch eine Verlangsamung der Durchblutung. Der tonischen und lange Zeit bestehenden Engerstellung der Gefäße folgen pathologische Veränderungen an den Gefäßwänden, die auch nach der Dekortikation und Wiederentfaltung der Lunge bestehenbleiben können. Beim starren Lungenkollaps wird die Mehrleistung des rechten Herzens unter Arbeit viel eher durch Drucksteigerung erreicht. Die Anwendung des Kollapses als therapeutisches Prinzip ergibt somit die eindeutige Bevorzugung solcher Methoden, die einen elastischen Kollaps hervorrufen. Beim plötzlich eintretenden Lungenkollaps nach chirurgischer Thoraxeröffnung konnten bemerkenswerte Drucksteigerungen im rechten herzen und der Arteria pulmonalis nicht nachgewiesen werden. Sie fanden sich aber, wenn unter intratrachealer künstlicher Beatmung größere Abschnitte der Lunge komprimiert wurden.
