    Description / Table of Contents: Zusammenfassung Bei Ratte, Maus, Meerschweinchen, Mensch, Kaninchen und Schwein konnte in vitro — und bei den beiden letztgenannten Species auch in vivo — eine lipolytische Wirkung eines von uns aus Schweinehypophysen dargestellten lipidmobilisierenden Polypeptids nachgewiesen werden. Beim Menschen zeigte sich nur ein Effekt auf die Glycerin-, nicht auf die Fettsäurefreisetzung; beim Schwein war in vitro keine Lipolysestimulation zu erzielen, in vivo dagegen eine starke Wirkung. Wie Untersuchungen der Cyclo-AMP-Phosphodiesterase-Aktivität im Fettgewebe des Menschen und der Ratte ergeben haben, ist als möglicher Wirkungsmechanismus eine Hemmung dieses Enzyms zu diskutieren. — Dieser Nachweis einer offenbar universellen Wirksamkeit stellt einen weiteren Hinweis auf das Vorhandensein eines speziellen lipidmobilisierenden Peptids in der Hypophyse dar.
    Notes: Summary The lipolytic effect of a polypeptide isolated from pig pituitaries was investigated in vitro and/or in vivo in 6 species. In vitro the peptide was active in the rat (MED 0,1 µg/ml), in the mouse and guinea-pig (MED 1,0 µg/ml each) and in the rabbit (MED 0,01 µg/ml). In human adipose tissue there was only an effect on glycerol liberation, in the pig no effect at all. But in vivo a strong action could be demonstrated in the minipig by injection of 100 µg/kg intravenously. Phosphodiesterase activity in adipose tissue of man and rat was inhibited by the peptide.

Weiterhin wird die Lipolyseaktivität von TSH und Peptid A bei der Ratte verglichen., Peptid A selbst hat biologisch einen TSH-Gehalt unter 25 mIE/mg. Um nachzuweisen, daß im TSH eine Peptid A-Verunreinigung vorliegt, wird säulenchromatographisch eine Fraktion TSH I/2 abgetrennt, die für die Kreuzreaktion und die lipolytische Wirksamkeit beim Kaninchen verantwortlich ist.
    Notes: Summary TSH from pig pituitaries (0.5 IU/mg) reacts with the antiserum of the lipid-mobilizing peptide A with an identical precipitation line. Lipolysis is stimulated in vivo and in vitro in rabbit adipose tissue. Furthermore, lipolytic activity of TSH and peptide A in the rat is compared. Peptide A itself biologically has a TSH-content of less than 25 mIU/mg. To show that TSH has a peptide A contamination, a fraction TSH I/2 is separated by column chromatography. TSH I/2 is responsible for the cross reaction and the lipolytic effectiveness in the rabbit.

    Description / Table of Contents: Zusammenfassung Innerhalb einer auf 3 Wochen begrenzten totalen Nahrungskarenz zeigten Haptoglobin, Transferrin und β1A-Globulin einen ausgeprägten, kontinuierlichen Abfall, das Complement lag am Ende deutlich unter dem Referenzbereich. Albumin, α2-Makroglobulin und Coeruloplasmin stiegen bis zum 10. Tag und fielen danach ab. Während sich bei den Immunglobulinen A und M ein Anstieg zeigte, blieb das IgG im wesentlichen unverändert. Wegen der beobachteten, erheblichen Veränderungen der Proteinfraktionen erscheint eine über den Zeitraum von 21 Tagen hinausgehende Nulldiät zumindest in dieser Form nicht gerechtfertigt.
    Notes: Summary During a period of total fasting restricted to three weeks, haptoglobin, transferrin and β1A-globin showed a marked, continuous decline. The complement was finally distinctly below the reference range. Macroglobulins and coeruloplasmin rised until the tenth day and decreased thereafter. IgG globulin remained essentially unchanged while immunoglobulins γA and γM showed an increase. On account of the considerable fluctuations observed in the protein fractions fasting in this form at least beyond a period of 21 days does not seem justified.

    Description / Table of Contents: Zusammenfassung Bei einer Patientin mit einer reticulocytogenen lipophagen Granulomatose in Unterhautfettgewebe und Lunge sowie zwei Patientinnen mit Mamma-Carcinom wurden im Tumorgewebe die Aktivitäten folgender Enzyme bestimmt: G-6-PDH, 6-PGDH, HK, PFK, PK, GDH, GK, MDH, GPM, GPT, GOT und GluDH. In Übereinstimmung mit der Literatur fanden wir in beiden Geweben ein tumorspezifisches Enzymmuster. In den Retikuloseknoten ließ sich darüberhinaus eine deutlich meßbare Aktivität der Glycerokinase nachweisen. Die „Michaelis-Konstante“ für Glycerin unterschied sich nicht von dem für Lebergewebe angegebenen Wert. Gut meßbare Aktivitäten dieses Enzyms sind bisher nur für Leber- und Nierengewebe angegeben worden.
    Notes: Summary The activities of the following enzymes were determined in a patient showing reticulo-cytogenous lipophagic granulomatosis in her subcutaneous adipose tissue and the lungs, as well as in two patients with carcinoma of the breast: G-6-PDH, 6-PGDH, HK, PFK, PK, GHD, GK, MDH, GPM, GPT, GOT and GluDH. We found a tumor-specific enzyme pattern in both tissues, a finding which is in accord with the results published in the literature. Furthermore, a well measurable glycerokinase activity in the reticulo-cytogenous lipophagic granulomas could be demonstrated as well. The Michaelis-Menten constant for glycerol was not different from the values found in hepatic tissue. So far, these enzymatic activities had been found to be easily assessable in liver and kidney tissue only.

    Description / Table of Contents: Zusammenfassung Im Humanserum sind bisher zwölf verschiedene Apolipoproteine nachgewiesen worden. Apo A–I and Apo A–II sind Strukturproteine der HDL und haben Coenzymfunktion für die LCAT. Apo B ist Hauptprotein der LDL, kommt aber auch in den triglyceridreichen Lipoproteinen vor und ist verantwortlich für die LDL-Rezeptorbindung. Die C-Apolipoproteine sind Oberflächenproteine der VLDL und werden bei deren Abbau auf die HDL übertragen; gleichzeitig haben sie spezifische Wirkungen auf die Lipoproteinlipase. Diese wird auch von den E-Apolipoproteinen beeinflußt, die sowohl in triglyceridreichen Lipoproteinen wie HDL vorkommen und für die Lipoproteinbindung an einen weiteren Zelloberflächenrezeptor verantwortlich sind. Ernährung und Medikamente, Hormone und Körpergewicht, Alkohol, Zigarettenrauchen und körperliches Training und schließlich Erkrankungen von Leber und Nieren beeinflussen Konzentration und Stoffwechsel der Apolipoproteine. Abschließend werden die Zusammenhänge mit der Arteriosklerose beim Menschen besprochen.
    Notes: Summary Twelve different apolipoproteins have been described in human serum. Apo A–I and apo A–II are essential for the structure of the HDL particles and for the function of LCAT activity. Apo B is the main protein in LDL but does also occur in the triglyceride-rich particles. Apo B represents the binding protein for the LDL-receptor pathway. The C-apolipoproteins are located on the surface of VLDL. They are transferred to HDL throughout the catabolism of VLDL and affect lipoprotein lipase activity. This enzyme is also affected by the E-apolipoproteins which occur in the triglyceride-rich particles as well as in HDL. Apo E is the binding site for another specific cell receptor. The concentration and metabolism of apolipoproteins is affected by diet, drugs, hormones, body weight, alcohol, cigarettes, physical exercises, liver and renal diseases. There is a close relation between apolipoproteins and atherosclerosis.

    Description / Table of Contents: Zusammenfassung Aus Hypophysen vom Schaf, Schwein, Rind und vom Menschen wurden zwei Polypeptide isoliert, die eine zum Teil ausgeprägte lipolytische Wirkung bei zahlreichen Spezies entfalten. Diese als Lipotropine bezeichneten Peptide wurden in 3 Fällen in ihrer Primärstruktur aufgeklärt; von weiteren liegen Angaben über die Aminosäurenzusammensetzung und erste Untersuchungen zur Sequenz vor. Danach unterscheiden sich die Lipotropine von den etablierten Hypophysenhormonen. Ihre Prüfung auf andere Hormonaktivitäten ergab bis auf MSH und zum Teil auch ACTH keine nennenswerten Aktivitäten; die Strukturgemeinsamkeiten zwischen den genannten Polypeptiden könnten eine Erklärung dafür sein. Erfolgsorgan ist das Fettgewebe, der Wirkungsmechanismus läuft über das Cyclo-AMP-System, wobei eine eindeutige Calciumabhängigkeit besteht. Es liegen Anhaltspunkte für eine gleichzeitige lipogenetische Wirkung der Lipotropine vor. An weiteren biologischen Wirkungen wurden Hyperglykämie, ein insulinotroper Effekt, Hypocalcämie und Gerinnungsaktivierung beschrieben. Mit Lipotropinen lassen sich spezifische Antikörper erzeugen, die in kleinen Konzentrationen die lipolytische Wirkung aufheben. Weiter konnte für β-LPH vom Schaf eine radioimmunologische Nachweismethode entwickelt werden, mit der beim Schaf eine Lipotropin-Serumkonzentration von 10 ng/ml festgestellt wurde. Erste Untersuchungen über die Lokalisation der lipolytischen, lipogenetischen und antigenen Aminosäurensequenz liegen vor. Für die Existenz eines speziellen lipidmobilisierenden Hormons sprechen also: 1 Die von den bekannten Hypophysenhormonen unterschiedliche Primärstruktur, 2 die spezifische radioimmunologische Nachweismethode, 3 das breite Speziesspektrum der lipolytischen Wirksamkeit, das sich von dem der anderen Hypophysenhormone unterscheidet.
    Notes: Summary Two lipid mobilizing peptides (lipotropins) which are lipolytically active in 10 species have been isolated from pituitaries of sheep, pig, bovine, and man. The amino acid composition of 15 lipotropins has been investigated, the primary structure is known in three of them. This demonstrates that the lipotropins are different from the known pituitary hormones. Containing the sequence of β-MSH, they have a strong melanocyte stimulating activity. Until now, one can only speculate whether adrenocorticotrophic activity, which differs from one to the other lipotropin, is due to contamination or to the primary structure because of the heptapeptide which they have in common with ACTH. Other pituitary hormone activities are minimal or absent. Via activation of adenylate cyclase and possibly inhibition of phosphodiesterase activity lipotropin is active in intact adipose tissue as well as in isolated fat cells. The stimulation of cyclo-AMP-system is calcium-dependent; in vivo hypocalcaemia and calcium ion influx into adipose tissue could be demonstrated. There is evidence for a simultaneous lipogenetic action of the lipotropin molecule. Further biological actions are hyperglycaemia, activation of the coagulation system, an insulinotropic and possibly glucagonotropic effect. In the rabbit, lipotropins induce specific antibodies which completely inhibit the lipolytic action of these polypeptides. For ovine β-lipotropin a sensitive and specific radioimmunoassay has been developed, showing a β-lipotropin concentration in sheep serum of 10 ng/ml. The following characteristics are in favour for the existence of a special lipid-mobilizing hormone: 1) the primary structure different from that of the known pituitary hormones, 2) the specific radioimmunoassay, 3) the wide species spectrum of lipolytic activity which is different from that of other pituitary hormones.

    Description / Table of Contents: Zusammenfassung Bei 12 Patienten mit primärer Typ IV-Hyperlipoproteinämie wurde der lipidsenkende Effekt konstanter Dosen der Biguanide Metformin, Phenformin und Buformin verglichen. Nach einer zweimonatigen Diätphase hatten sich Körpergewicht, Triglyceridausgangswerte und Cholesterinplasmakonzentrationen stabilisiert. Die 3 Biguanide wurden jeweils über 2 Monate gegeben. Auf diese sechsmonatige Medikamentenperiode folgte eine weitere Diätphase. Während 0,15 g Phenformin und 2,55 g Metformin die Triglyceride um ca. 50% senkten, betrug die Senkung unter 0,30 g Buformin nur die Hälfte. Dies ist zurückzuführen auf 4 Therapieversager mit Triglyceridausgangswerten von über 700 mg/100 ml. Im Gegensatz dazu war der triglyceridsenkende Effekt der beiden anderen Biguanide umso stärker, je höher die Triglyceridausgangswerte waren. Der cholesterinsenkende Effekt der Biguanidbehandlung war weniger stark ausgeprägt. Signifikante Veränderungen des Körpergewichts sowie ein Einfluß auf Basalinsulinwerte und Glucosekonzentrationen konnten nicht beobachtet werden. Alle 3 geprüften Biguanide bewirken also in einer Dosis von jeweils 3 Tabletten eine Senkung der Triglyceride, obwohl sie in ihrer Wirksamkeit unterschiedlich und vom Grad der Type IV-Hyperlipoproteinämie abhängig sind.
    Notes: Summary In 12 patients with primary type IV hyperlipoproteinemia the lipid lowering effect of constant doses of the biguanides metformin, phenformin and buformin was compared. After a 2 months dietary period bodyweight and initial triglyceride and cholesterol plasma concentrations were stabilized. The three biguanides were given for two months each, and this 6 months drug period was followed by another dietary period. While, 0.15 g phenformin and 2.55 g metformin reduced triglycerides by about 50% the reduction by 0.30 g buformin was only half. This was due to 4 nonresponders with initial triglycerides of more than 700 mg/100 ml. In contrast the triglyceride lowering effect of the other two biguanides was positively correlated to the pretreatment triglycerides. The cholesterol lowering effect of biguanide treatment was far less pronounced. There were no significant alterations of body weight and no influence on basal insulin and glucose concentrations. In conclusion, all three biguanides tested in a dosis of 3 tablets each are effective in lowering triglycerides although differing in efficiency and depending from the degree of typ IV hyperlipoproteinemia.

    Description / Table of Contents: Zusammenfassung Der Einfluß einer vierwöchigen Clofibratbehandlung auf den Triglycerid-, Cholesterin-und Phospholipidgehalt von Very Low Density Lipoproteins (VLDL), Low Density Lipoproteins (LDL) und High Density Lipoproteins (HDL) wurde bei 12 Patienten mit primärer Typ IV-Hyperlipoproteinämie untersucht. Die Senkung der Serumtriglyceride war durch einen Abfall der VLDL-Triglyceride bedingt. Darüber hinaus kam es zu einer Normalisierung der zum Teil nachweisbaren LDL-Hypertriglyceridämie. — Die Cholesterinkonzentration nahm in den VLDL und HDL, der Phospholipidgehalt nur in den VLDL signifikant ab. In den LDL änderten sich diese beiden Lipide in Abhängigkeit von ihren Ausgangswerten, die negativ mit den Serumtriglyceridkonzentrationen korrelierten. Die Normalisierung von VLDL- und LDL-Lipiden könnte mit einer beschleunigten Delipidation der Apo-B-Lipoproteine erklärt werden.
    Notes: Summary The effect of clofibrate treatment (1.5 g/day for four weeks) on triglycerides, cholesterol and phospholipids in very low density lipoproteins (VLDL), low density lipoproteins (LDL) and high density lipoproteins (HDL) was investigated in 12 patients with type IV hyperlipoproteinaemia. The decrease of serum triglycerides was due to the reduction of VLDL-triglycerides. Furthermore, LDL-hypertriglyceridemia was normalised by clofibrate. — There existed a significant decrease of VLDL- und HDL-cholesterol as well as of VLDL-phospholipids. The changes in LDL-cholesterol and LDL-phospholipids by clofibrate depended on their initial concentrations which were negatively correlated to the serum triglycerides before treatment. — The normalisation of lipid concentrations in VLDL and LDI could be explained by a stimulated delipidation of the apo B-lipoproteins.

    Description / Table of Contents: Summary The lipids in normal human Leukocytes — isolated by a method described previously — were determined by microliter-methods. We found the following average values per 109 cells: total lipids 15,56 mg, lipidphosphorus 0,42 mg, phospholipids 10,43 mg, total cholesterol 2,83 mg, unesterified cholesterol 2,32 mg and total esterified fatty acids 6,48 mg.
    Notes: Zusammenfassung Es wird über Lipidbestimmungen in normalen menschlichen Leukocyten mit Mikrolitermethoden und einem früher beschriebenen Leukocytenisolierungsverfahren berichtet. Dabei fanden sich als Mittelwerte pro 109 Zellen: 15,56 mg Gesamtlipide, 0,42 mg Lipoidphosphor entsprechend 10,43 mg Phospholipoiden, 2,83 mg Gesamtcholesterin, 2,32 mg freies Cholesterin und 6,48 mg Esterfettsäure.
