Notes: Abstract. Objective: To examine the effect of hyaluronic acid (HA) on the induction of superoxide anion by IL-1 in chondrocytes. ¶Materials and Methods: Bovine articular chondrocytes were treated with different concentrations of IL-1. A chemiluminescent probe (L-012) was added to the medium and chemiluminescence detection was used to measure superoxide anion. ¶Results: IL-1 caused induction of superoxide anions in a dose-dependent manner. HA (10–100 〈mu〉g/ml) countered superoxide induction caused by 20 ng/ml of IL-1. ¶Conclusion: HA can afford protection against cartilage degradation, probably acting as a free-radical scavenger.

Notes: Abstract. Objective: To examine the effect of hydrogen peroxide on chondrocyte metabolism.¶Materials and Methods: Bovine articular chondrocytes were used. Proteoglycan (PG) synthesis was measured with [35S] sulfate incorporation. For detection of apoptosis, the TdT-mediated dUTP-biotin nick end labeling (TUNEL) and annexin V assay were used. Extracellular-regulated protein kinase (ERK) activity was measured using a mitogen-activated protein kinase assay system.¶Results: Addition of hydrogen peroxide resulted in the inhibition of PG synthesis, apoptosis, and enhanced ERK activity.¶Conclusion: Hydrogen peroxide plays an important role in regulating the metabolism of chondrocytes.

Notes: Abstract We investigated the importance of prostaglandin E2 (PGE2) release in interleukin-1 (IL-1)-induced inhibition of aggrecan synthesis by chondrocytes. Keratan sulfate (KS) production was measured in parallel with PGE2 release in chondrocytes. IL-1 inhibited KS production and stimulated PGE2 release. In the presence of PGE2, there was a dosedependent decrease in baseline KS production. Indomethacin and dexamethasone partially blocked the IL-1-induced PGE2 release while KS production recovered. Our results suggest that IL-1 inhibits KS production, in part, by stimulating the release of PGE2.

Notes: Summary Forty-two cadaver knees were used for morphologic and MRI observations of the tendinous distal expansions of the semimembranosus m. and the posterior capsular structures of the knee. A tendinous branch of the semimembranosus m. inserting into the posterior horn of the lateral meniscus was found in 43.2% of the knees dissected, besides five already known insertional branches; capsular, direct, anterior and inferior, as well as the oblique popliteal ligament. The tendon had three morphologic types; thin, broad and round. All three types moved the lateral meniscus posteriorly when pulled on. Thus, the semimembranosus m. may also have a protective function for the lateral meniscus as well as the already well established function of protecting the medial meniscus in knee flexion. When a semimembranosus tendon attachment to the posterior horn of the lateral meniscus is present, its normal insertion is difficult to differentiate from a lateral meniscus tear in MRI and this may cause misdiagnosis.

Notes: Abstract We found a left superficial ulnar artery in the cadaver of a Japanese woman. This anomalous vessel originated from the brachial artery at a site 55 mm distal to the inferior border of the teres major muscle and medial to the median nerve, ran downward and medially superficial to the forearm flexor muscles, and then downward to enter the hand. It formed superficial and deep palmar arches with the radial artery. The clinical importance of the anomalous ulnar artery is discussed.

Notes: Summary During diapause O2 consumption in fly pupae is a cyclic event (4-day periodicity at 25°C) driven by cycles of juvenile hormone activity. Levels of juvenile hormone esterase activity change systematically during the cycle, with highest activity observed at the nadir of the O2 consumption cycle.

Notes: Summary Plasma leptin level is known to correlate with the degree of obesity. To determine the influences of renal fuction and insulin resistance on plasma leptin concentrations, we measured plasma leptin concentrations and performed the euglycaemic hyperinsulinaemic clamp studies in 57 patients with non-insulin-dependent diabetes mellitus with a wide range of renal function. In simple regression analyses, plasma leptin concentration showed significant positive correlations with percentage of body fat measured by dual energy X-ray absorptiometry, body mass index, waist to hip ratio and fasting plasma insulin. Leptin level was higher in females than males. Multiple regression analyses indicated that percent body fat, waist to hip ratio, plasma insulin, gender and renal function (1/creatinine), but not insulin sensitivity, were significant and independent determinants of plasma leptin level. These results suggest that plasma leptin level is regulated or affected by multiple factors including renal function. Insulin resistance appeared to increase leptin levels indirectly by raising plasma insulin. [Diabetologia (1997) 40: 676–679]
