    Notes: Abstract The extent to which the plasticity in peptide expression observed in developing spinal motoneurons occurs following proximal peripheral axotomy in the adult rat was examined using in situ hybridization and immunohistochemical techniques to visualize the changes. Transient upregulation of galanin, vasoactive intestinal polypeptide (VIP) and substance P messenger ribonucleic acids (mRNAs) was observed within subpopulations of motoneurons ipsilateral to lesion for periods lasting 2–3 weeks after injury. In contrast, the axotomy-induced heterogenous increases in somatostatin and neuropeptide tyrosine mRNA expression in ipsilateral motoneurons remained elevated, or, in the case of somatostatin, continued to increase for the time period studied (1 month). Immunohistochemical analysis agreed with the in situ hybridization results, showing some motoneurons within the injured ventral horn to contain galanin-, VIP-or somatostatin-like immunoreactivity. In some instances, galanin-immunoreactive motoneurons colocalized with calcitonin gene-related peptide immunoreactivity. Most of the neurons expressing the injury-induced peptides appeared large, presumably alpha-motoneurons, but there were also many small neurons expressing galanin in the ventral horn ipsilateral to lesion. This may represent evidence for peptide synthesis in gamma-motoneurons. The only peptide mRNA studied to be downregulated in response to axotomy was enkephalin. The results show that peptide expression in injured motoneurons is dramatically altered, the significance of which remains to be determined.

    Notes: Summary In the present study we examined the effect of systemic tocainide on sensory hypersensitivity in rats after spinal cord ischemia induced by a photochemical technique. After induction of spinal cord ischemia the rats exhibited a sensory disturbance which was mainly expressed as vocalization to innocuous cutaneous mechanical stimuli (allodynia) in the flank area during the following several days. Tocainide at 75 mg/kg i.p., but not 50 mg/kg i.p., significantly increased the vocalization threshold to mechanical pressure for 2 h. The effect of intraarterial (i.a.) tocainide on the responses of dorsal horn wide-dynamic-range (WDR) neurons to suprathreshold electrical stimulation of their receptive fields was also examined in normal rats and after transient spinal cord ischemia, at a time when the animals exhibited typical behavioral allodynia in the dermatomes innervated by the ischemic spinal segments. In normal rats, tocainide (50 mg/kg i.a.) strongly suppressed the responses of WDR neurons to C fiber input with lesser effect on A fiber input. In allodynic rats, tocainide suppressed the augmented A and C fiber mediated responses of WDR neurons to the extent that their responses were similar to those seen in normal rats without tocainide. There was no difference in the overall depression of A and C fiber mediated input by tocainide between normal and allodynic rats. The present results demonstrated the analgesic effect of systemic tocainide in relieving allodynia in rats and indicated that systemic local anesthetics, at doses that do not block nerve conduction, can be effective in suppressing dorsal horn WDR neuronal activity. Although such drugs primarily suppress C fiber induced activity, the depression by local anesthetics of increased A fiber induced responses in allodynic conditions mediated by myelinated afferents may explain the analgesic effect of such drugs on behavior.

    Notes: Summary The capacity of the saphenous nerve for collateral sprouting was examined by electrophysiological recordings of the activity of low threshold mechanoreceptors and plasma extravasation after C-fiber stimulation in rats. When the sciatic nerve was sectioned neonatally or in adults little evidence was obtained for collateral sprouting of either mechanoreceptors or fibers involved in plasma extravasation in the intact saphenous nerve0 In rats where the sciatic nerve was sectioned and the saphenous nerve was crushed either neonatally or in adults, expansive regenerative reinnervation by thin fibers, but not mechanoreceptors, was observed particularly in glabrous skin. Saphenous crush alone did not cause expansive regenerative reinnervation. The results indicate that much of the collateral sprouting or regenerative reinnervation of the skin observed in morphological studies may represent the presence of fibers not responding to the stimuli used in the present study. It is also suggested that some observations of collateral sprouting may represent changes in responsiveness to stimulation of skin areas with overlapping innervation territories.

    Notes: Summary The neuropeptide galanin (GAL) was injected intrathecally (i.t.) in decerebrate, spinalized, unanesthetized rats and its effect on the nocifensive flexor reflex was examined. The reflex, which was evoked by intense mechanical or thermal stimulation of the foot, was recorded from the ipsilateral hamstring muscles. I.t. GAL increased reflex excitability significantly more to thermal than to mechanical stimuli. It is suggested that GAL, which is present in sensory fibers that innervate the skin, is released by the central terminals of cutaneous afferents that are much more sensitive to thermal than to mechanical stimuli.

    Notes: Abstract Two sets of experiments were carried out in order to evaluate the unmyelinated (C) fibre innervation of the hindlimb of rats. In the first experiment the normal innervation territory of the various nerves to the skin in the hindfoot was examined by plasma extravasation of Evans blue after antidromic nerve stimulation. Considerable overlap was found in the territories of nerves on the dorsum of the toes and just proximal to the toes. In the second experiment the capacity of C-fibres in the saphenous nerve for growth into deafferented skin was examined. In rats in which the sciatic nerve was previously sectioned, the innervation territory of the intact saphenous nerve expanded slightly into that of the sciatic in about 20% of the cases by “collateral sprouting”. When the sciatic nerve section was combined with ipsilateral saphenous nerve crush, the regenerating C-fibres in the saphenous nerve innervated the sciatic territory in almost 100% of cases, a process we termed “expansive regenerative reinnervation”. We conclude that the existence of overlapping C-fibre innervation territories need to be considered in studies of “collateral sprouting”. However, regenerating C-fibres can innervate foreign deafferented skin.
