    Notizen: Circular magnetic x-ray dichroism is a promising new tool for the study of spin and orbital moments in ferromagnetically ordered materials. For rare-earth systems the L2 and L3 edge dichroic spectra give information about the magnetic properties of the 5d conduction electrons. This article addresses a few of the key issues concerning the interpretation of the spectra. In particular, the quadrupole nature of the dichroic spectra below the L absorption edges and its angular dependence is discussed. The important role of the 4f-5d exchange in causing a large and important spin dependence in the 2p–5d matrix elements is likewise examined.

    Notizen: Laterally gain-coupled distributed feedback GaInAsP/InP lasers were fabricated by focused ion-beam implantation and rapid thermal annealing of ridge waveguide structures. An absorption grating, which interacts with the evanescent light field, is formed by a periodic reduction of the band-gap absorption in the passive sections along the ridge. Room-temperature single-mode emission was observed at 1.5 μm with a 30 dB side-mode suppression ratio. Simultaneously, the increase of the band gap in the implanted section results in improved carrier confinement, which reduces the leakage current and improves the laser performance in comparison to unimplanted ridge waveguide lasers. © 1998 American Institute of Physics.

    Notizen: Complex coupled GaInAsP/InP distributed feedback lasers were developed based on maskless focused ion beam lithography. By combining implantation enhanced wet chemical etching and implantation induced thermal quantum well intermixing a refractive index grating was defined self-aligned to a gain grating forming a complex coupled grating lateral to a ridge waveguide. The devices show single mode emission at wavelengths around 1.55 μm with linewidths 〈2 MHz and side mode suppression ratios of more than 40 dB for continuous wave operation at room temperature. A high single mode yield (〉90%) over a large tuning range (88 nm) was achieved. © 1999 American Institute of Physics.

    Notizen: Abstract: Four main molecular forms of acetylcholinesterase (AChE) characterized by their sedimentation coefficients (5S, 7.5S, 11.5S, and 20S), are found in chick ciliary ganglion. After transection of the preganglionic nerve (denervation), total AChE activity in the ganglion dropped by 35% in 2 days. By then, 11.5s and 20s forms had diminished by 60 and 75% respectively, where as 7.5s remained practically unchanged. Since presynaptic structures disappeared 2 days after denervation, we inferred that at most 35% of total ganglion AChE was presynaptic: 11.5s and 20s might be mainly presynaptic and 7.5S, postsynaptic. At later time intervals. total AChE continued to decline up to day 5, possibly as a result of orthograde transynaptic regulation of the enzyme activity. After transection of postganglionic nerves (axotomy), total ganglion activity showed little change; 11.5s and 20s decreased by 40 and 6076, respectively, in 5 days, but these drops were compensated for by an early increase in 7 5S, which started the day after axotomy. After simultaneous transection of both pre- and postganglionic nerves (double section), total ganglion AChE dropped rapidly by 35% in 1 day and remained at that level up to 21 days. The 11.5S diminished rapidly by 60% in 1 day. The early increase of the 7.5s form induced by axotomy alone did not occur. Since the effect resulting from double section was not the equivalent of the cumulative effects observed after denervation and axotomy, respectively, the level of AChE forms in the ganglion may be regulated by reciprocal interaction of pre- and postsynaptic elements. After denervation and double section but not after axotomy alone, the contralateral non-operated ganglion exhibited a fall in the 20s form. This suggests that a transynaptic effect is exerted on AChE by the contralateral preganglionic neuron. Taken together, these results indicate that the various AChE molecular forms in chick ciliary ganglion are preferentially but not exclusively distributed as follows: the pre- and postganglionic axons contain mainly the 11.5S form, whereas nerve endings and synaptic structures are enriched in 20S, and ganglion cell bodies, in 7.5s.

    Notizen: —Highly purified fractions of synaptic vesicles were prepared from rat cerebrum or cerebral cortex by density gradient centrifugation. Treatment of synaptic vesicle fractions by autoincubation, freeze-thawing and sonication in an isotonic alkaline-salt medium or in 0·1-0·3% (v/v) Triton X-100 released increasing quantities of synaptic vesicle protein and phospholipid into solution. When the soluble synaptic vesicle proteins were extracted with 0·1% (v/v) Triton X-100, the insoluble residue consisted mostly of 5–8 nm-thick membranes resembling the limiting membranes of intact synaptic vesicles. This finding, together with other considerations, suggested that the soluble proteins and accompanying phospholipids originated from the interior of the synaptic vesicles. A 0·3% (v/v) Triton X-100 extract of synaptic vesicle was fractionated by ultracentrifugal flotation and dialysis into three lipoprotein fractions: a low density lipoprotein (d 〈 1·21 g/ml), a high density lipoprotein (d = 1·21–1·35 g/ml) and a very high density lipoprotein (d 〉 1·35 g/ml). The phospholipid contents of the low, high and very high density lipoprotein fractions were 0·74, 0·38 and 0·20 mg/mg of protein, respectively. All three apolipoproteins had a high ratio of acidic to basic, and of polar to nonpolar, amino acids, and were rich in glycine, alanine and serine. Polyacrylamide gel electrophoresis of the alkaline-salt and Triton X-100 extracts of synaptic vesicles at pH 8·8 resolved a single anionic component which stained for protein, lipid (Sudan black B; iodine) and anionic groups (acridine orange). Polyacrylamide gel electrophoresis of synaptic vesicle extracts at pH 2·7 in 5 m urea and 0·25% (v/v) Triton X-100 resolved about 20 protein components. However, the protein profiles of electropherograms of the Triton X-100 and alkaline-salt extracts differed in certain respects, suggesting that these media to some extent solubilized different proteins. However, most of the protein bands in electropherograms of the Triton X-100 and alkaline-salt extracts also stained for lipid and anionic groups. In addition, two lipoprotein components in the alkaline-salt extract and four in the Triton X-100 extract contained carbohydrate. Isoelectric focusing of synaptic vesicle extracts resolved 6–8 protein fractions. The major fraction in Triton X-100 and alkaline-salt extracts had an apparent isoelectric point of approximately 4·2 and contained 0·24 mg of phospholipid per mg of protein. Soluble synaptic vesicle proteins released by incubating, freeze-thawing and sonicating in the alkaline-salt medium, and protein fractions of the latter obtained by electrofocusing had an absorption maximum of 260–265 nm which was enhanced in a cold 0·5 n perchloric acid extract, an observation suggesting the presence of a bound nucleotide. These findings demonstrate that rat brain synaptic vesicles contain a heterogenous array of soluble acidic lipoproteins which vary in buoyant density, lipid content, amino acid and carbohydrate composition and electrophoretic mobility in polyacrylamide gels. These acidic lipoproteins apparently comprise the bulk of the macromolecular contents of synaptic vesicles and probably serve as ‘carrier’ proteins for the binding and sequestration of the neurotransmitters.

    Notizen: Abstract— Some metabolic and biochemical effects of fluorocitrate were studied in vivo in rat brain and cat spinal cord. During the preconvulsant and convulsant phases of fluorocitrate poisoning the contents of free glutamate, glutamine and aspartate declined progressively, while that of alanine increased. Incorporation of 14C from [U-14C]glucose into these amino acids also decreased, although somewhat more gradually. GABA exhibited a biphasic change, its content rising after an initial decrease while its relative specific activity rose initially and subsequently diminished. Incorporation of 14C from [U-14C]glucose and [U-14C]lysine into neural protein declined sharply. The citric acid content rose markedly in rat brain and cat spinal cord. In rat brain the glycogen content declined but ATP and ammonia contents were unchanged. The significance of these results with respect to energy metabolism and the possible mechanism of the convulsions during fluorocitrate poisoning is discussed.

    Notizen: The de novo biosynthesis of purine nucleotides in rat brain was markedly and rapidly inhibited by intracerebral injections of azaserine. After 48 hr the animals became paraplegic. The biosynthesis of purine and pyrimidine nucleotides, RNA and protein was investigated by radioactive tracer studies over a 4-day period in an attempt to correlate the azaserine-induced, metabolic lesions found in rat brain and the manifested, peripheral, neurological disturbance.
