Notizen: Objective: The aim of this study was to examine the association between medication exposure and (1) unstimulated whole-salivary flow rate and (2) the severity of xerostomia among older people while adjusting for multiple medication use. Methods: Data were obtained from participants remaining at the five-year follow-up phase of a cohort study of community-dwelling older South Australians. Medication exposure information was available at baseline and at five years, enabling examination of the effects on dry mouth of long-term exposure to medications. At the five-year follow-up, unstimulated salivary flow was estimated using the spit method, and xerostomia severity was estimated using the 11-item Xerostomia Inventory. Because of the potential difficulties posed by polypharmacy, a two-stage analytical approach was employed: (1) Classification and Regression Tree (CART) analysis was used as an exploratory device to elucidate the relationships among the dependent and independent variables, and (2) linear regression analysis was used as a complementary procedure. Results: Unstimulated flow rate was lower among individuals who were female or taking antide-pressants at both baseline and five years, and higher among smokers or people who were taking hypolipidemic drugs. Xerostomia severity was higher among females, or individuals taking: (1) an anginal at baseline and five years, (2) an anginal without a concomitant betablocker at five years, (3) thyroxine and a diuretic at five years, or (4) antidepressants or antiasthma drugs at both baseline and at five years. Conclusions: These results suggest that polypharmacy can be accounted for to a certain extent by using CART analysis in conjunction with more conventional approaches; and that the relationship between medications and dry mouth is a complex one, and differs according to which aspect of dry mouth is being examined.

Notizen: We have characterized TAP allele frequencies in a panel of 71 Yoruba Nigerians using ARMS-PCR. With the exception that TAP 2D was absent in Nigerians, TAP 2 allele frequencies in this population were found to be similar to those in a UK white population. HLA-DR4 also was found to be at a low frequency in Yoruba Nigerians (1.4%). This may reflect the absence of TAP 2D in Nigerians as DR4 and TAP 2D are in linkage disequilibrium in UK Caucasoids. The most frequent TAP 1 allele in Yoruba Nigerians was TAP 1A (49%). However, this value will be an underestimate as TAP1 alleles could not be unequivocally assigned in 41 % of subjects using the ARMS-PCR methodology.

Notizen: An accurate and precise method was developed for the determination of carrageenan in milk-based infant formulas. Following digestion of protein with papain, carrageenan was precipitated with Hyamine 1622, a quatemary ammonium salt. The precipitate was washed to remove lactose, dissolved in sulfuric acid and the carrageenan assayed by color development with the phenol-sulfuric reagent. Filtrations and washings were facilitated with Millipore filters. Carrageenan recovery of spiked-formulas averaged 99.67% for ready-to-feed products and 95.74% for concentrated products, which respectively contained 7.1% and 14.2% lactose. To obtain these accuracies the reference carrageenan must be identical to the carrageenan in the product.

Notizen: The caustic peeling rates of potatoes is considered from an analytical and experimental point of view. The analysis considers the process to be governed by the competing rates of mass transport within the potato (diffusion) and chemical reaction. The chemical reaction between caustic and chlorogenic acid is singled out for analysis since the product of this reaction is a yellow salt which is easily measured by calorimetry. Simplified equations for the concentration of the yellow salt as a function of time is obtained for the two limiting extremes of diffusion controlled rates and reaction controlled rates. Experiments on Idaho Russetts indicate that, over the first 2–3 min of immersion in caustic, the process is diffusion controlled. It is also shown that it is the outer skin which presents the large diffusion resistance. For reasonable times after this initial period the process is reaction controlled and very sensitive to temperature. The implications of these results with respect to processing techniques (e.g., the dry peel process) and sequences is considered and discussed.

Notizen: The use of the new Medishield anasthetic gas scavenging system with ventilators in the Siemens-Elema Servo 900 series was investigated. It was discovered that this combination of equipment creates the hazard that a disconnexion in the patient breathing system may not be detected by the ventilator alarm. This happens because the suction generated by the scavenging system draws a considerable flow of room air through the expiratory system of the ventilator when a disconnexion occurs. Possible solutions to the problem were investigated, and it was discovered that the problem can be overcome by applying a small amount of positive end-expiratory pressure to counteract the suction from the scavenging system.
