    Notizen: Abstract: Melittin enhanced sphingomyelin (SPM) degradation by the neutral membrane-bound sphingomyelinase from calf brain synaptosomal plasma membranes (SYM) up to 20-fold. Melittin in concentrations as high as 100 μM did not significantly alter membrane fluidity of SYM as measured by fluorescence depolarization and electron spin resonance (ESR) using diphenylhexatriene and a doxy1 derivative of SPM, respectively. In the concentration range 100-1000 μM. melittin was observed to rigidify SYM. The incorporation of SPM.erivatives into the lipid bilayer of SYM.as demonstrated by ESR measurements. Melittin enhanced the uptake of SPM-derivatives into SYM.

    Notizen: The total N-acetyl-β-d-hexosaminidase (hexosaminidase A plus B) activity of the human brain increases during life until the age of 70 years to about twice the activity found in the foetal brain. This activity roughly parallels the increasing level of brain ganglio-sides. Simultaneously, the ratio between the hexosaminidases A and B changes from approx. 3.4 to 1.1 as established by the microscale isoelectric focussing of the enzymes from crude extracts. Age-dependent variations can be also demonstrated for other glycosidase activities: The β-d-glucosidase is shown to increase, the α-d-mannosidase and α-l-fucosidase to decrease during life. The enzyme values are related to the protein content, the age-dependent shift of which we have determined in the human brain. In different organs and body liquids distinct differences are found for the levels of the total hexosaminidase activity as well as for the hexosaminidase pattern. The relation of the hexosaminidase activity to the ganglioside degradation during the human development is discussed.

    Notizen: Abstract: High concentrations of halothane stimulate the degradation of sphingomyelin by neutral sphingomyelinase bound to synaptosomal plasma membranes (SYM) from calf brain up to 50-fold, and increase membrane fluidity dramatically as measured by fluorescence depolarisation of incorporated 1,6-diphenylhexatriene (DPH). To investigate the effects of low concentrations of halothane clinical conditions of anaesthesia were simulated in vitro by gassing a suspension of SYM with a gas mixture of 1.5 and 3% (by vol.) of [14C]halothane in N2 at 37°C. The uptake of halothane into SYM as determined by radioactivity measurements was 31 and 73 mmol per mol membrane lipid or 4 and 9 mg per g membrane, respectively. The same concentrations of halothane in the membrane suspensions increased membrane fluidity of SYM significantly as shown by measurements of fluorescence depolarisation of in-corporated DPH. At halothane concentrations corresponding to 1.5 and 3% (by vol.) in the gas phase the degradation of sphingomyelin by membrane-bound neutral sphingomyelinase of SYM was stimulated by 11 and 57% of controls, respectively. These results are discussed in relation to the anaesthetic potential of halothane.     Notizen: In autopsy tissues of 12 cases of Tay-Sachs disease the N-acetyl-β-hexosamini-dase A and B activities were investigated using chromogenic and physiological substrates.In three cases of Tay-Sachs disease, classified as the variant O, the enzyme activities A and B were missing; in eight cases, classified as the variant B, the enzyme activity A was missing. In another case, both enzyme activities wcre shown to be enhanced in brain tissue (‘variant AB’), using a chromogenic substrate.The three enzymic variants showed different glycolipid storage patterns of Tay-Sachs-ganglioside (TSG) and its asialo residue, the trihexosylceramide (THC) in the nervous tissues. Additional storage of kidney globosidc was found in the visceral tissues of the O variant. A decrease of the non-accumulated lipids, especially of those characteristic for myelin, was observed. The quantitative lipid determinations were performed by means of a thin-layer densitometric micromethod (standard deviation 2–5 per cent).Evidence is presented that the different storage patterns result from the corresponding enzyme alterations in the three variants. An essential condition for this statement was the isolation of the storage compounds from Tay-Sachs tissues and their radioactive labelling by the addition of tritium to the double bond in their sphingosine moiety. In a previous investigation it was shown that enzyme A degrades the storage compounds TSG, THC and kidney globoside while enzyme B acts on THC and kidney globoside only. In agreement with this finding, a highly concentrated mixture of both enzymes from normal tissues hydrolyses the main storage compound, the Tay-Sachs-ganglioside. This hydrolysis was reduced when corresponding enzyme preparations from tissues of variants of Tay-Sachs disease (including variant AB) acted on TaySachs ganglioside.Some properties of the N-acetyl-β-D-hexosaminidases from normal and from pathological tissues were determined with chromogenic and physiological substrates. The relationship between the enzymes A and B is discussed.

    Notizen: Abstract: In contrast to extracellular, long chain ceramides which comprise a structural component of the epidermal water barrier, intracellular ceramides originating from sphingomyelin hydrolysis have been shown to inhibit proliferation and to induce apoptosis in different cell populations. To further elucidate the possible role of intracellular ceramides in human epidermis, two new cell-permeable ceramide analogues, N-thioacetylsphingosine (C2-Cer=S) and 4-dodecanoylamino-decan-5-ol (FS-5), were synthesized and tested for their ability to suppress cell growth and to induce apoptosis in immortalized human keratinocytes. It was shown that the well-investigated ceramide analogue N-acetylsphingosine (C2-Cer=O), as well as the new compound C2-Cer=S inhibited proliferation of HaCaT cells with half-inhibitory concentrations (IC50) of 20 μg/ml and 10 μg/ml, respectively, whereas FS-5 has been potent with an IC50〉40 μg/ml. Overall, all three ceramide analogues induced apoptosis in HaCaT cells as assessed by DNA-fragmentation using ELISA technique and in situ nick end labelling, thereby confirming the importance of ceramide signalling in keratinocytes.
