    Notes: Abstract: We have shown by northern analyses that the expression of the mouse polyubiquitin C gene is increased several fold in the brains of mice infected with both the ME7 and 87V strains of scrape. Expression of the polyubiquitin gene does not change significantly, compared with controls, until the later stages of disease progression when there is a 2.5-fold increase in ME7-infected brains and a 1.8-fold increase in 87V-infected brains. The patterns of changes of expression of the polyubiquitin genes in brains infected with the two strains of scrapie resemble those of accumulation of ubiquitin-conjugate-positive structures in the brain that are detected immunohisto chemically. A similar increase in the expression of a heatshock protein 70 gene also occurs.

    Notes: This study investigated the direct imaging of low intensity x-rays by a room-temperature charge coupled device (CCD). Commercial CCD evaluation kits read out the information and provided initial processing of the data. The extraction of the signal from noise was achieved using NIM modules rather than by storing and numerically analyzing the data. Due to the fast processing at standard TV rates, the storage of the data required conversion of the signal from a timing to a pulse height analysis (PHA) spectrum. The tested CCDs were found to have lower efficiency relative to the proportional counter for the range of photon energies (1.5–8.0 keV) studied but had higher spatial resolution than gas-filled position-sensitive detectors. Application of CCDs for x-ray energy dispersive detectors is described.

    Notes: We have recently built a spectrometer that efficiently collects the energy spectrum of charged particles that emerge from targets bombarded by low-flux (10−15 A) incident beams. The spectrometer is used to detect positron-induced secondary electrons and low-energy positron-stimulated Auger electron emission. Slow positrons and positron-induced secondary electrons are generated from a cold neon positron moderator deposited onto an encapsulated 22Na source. The moderated beam is magnetically transported to the target with two sets of E×B plates. The particles emerging from the target are energy dispersed by a third set of plates and are detected with a one-dimensional position-sensitive detector. The data-collection rate for the energy-distribution spectrum is ∼10–100 times faster for this experiment relative to the more common method of sweeping grids or the plates.

    Notes: We have used photocurrent measurements to demonstrate a strong electroabsorption effect in GexSi1−x/Si multiple quantum wells grown by remote plasma-enhanced chemical vapor deposition. Large voltage-induced shifts in absorption are observed at room temperature in the wavelength range from 1.2 to 1.58 μm. We anticipate that the results can be extended to fabricate GexSi1−x optoelectronic devices operating at room temperature.

    Notes: Abstract: The distribution of 5′-nucleotidase activity, dopaminergic [3H]spiperone binding sites, and [3H]quinuclidinyl benzilate (QNB) binding sites in different subcellular fractions of bovine caudate nucleus has been studied. Each activity was enriched in a microsomal (P3) preparation from that tissue. The microsomal preparation was further fractionated by different techniques. First, the P3 fraction, or a sonicated P3 fraction, was fractionated on a discontinuous sucrose density gradient. Second, the P3 fraction, or a digitonin pretreated P3 fraction, was fractionated on a continuous sucrose density gradient. The results obtained demonstrate that 5′-nucleotidase activity does not cofractionate with radioligand binding activity, although no difference between the distributions of [3H]spiperone binding and [3H]QNB binding were seen. It is concluded that the two radioligand binding activities are located on nonglial membranes.

    Notes: Abstract— [3H]Spiperone binding has been used to study neurotransmitter receptors in bovine caudate nucleus in displacement and saturation binding experiments. Displacement curves for several antagonists are biphasic and can be analysed into contributions from dopaminergic and serotonergic sites. Antagonist binding at each class of sites follows the simple mass action equations for binding at a homogeneous set of sites (slope factors close to unity). Agonist displacement curves also indicate complex behaviour, but agonist binding to the dopaminergic sites alone exhibits heterogeneous properties (slope factors less than unity). Saturation binding experiments have been conducted on each class of site, defining dopaminergic binding of [3H]spiperone as that binding displaced by 0.1 mm-dopamine and serotonergic binding as that displaced by 0.3 μm-mianserin. In each case, a single class of binding sites was detected: the binding parameters derived in this way have been used to calculate the proportions of the two classes of binding site observed in displacement experiments. Good agreement was obtained between calculated and observed values.

    Notes: The activities of mitochondrial hexokinase and adenylate kinase have been measured in various osmotic conditions. Sucrose, potassium chloride and ammonium acetate were used as solutes. The total hexokinase activity of mitochondrial suspensions increased steadily with decreasing osmolarity of the sucrose or salt solutions. The hexokinase activity of mitochondrial suspensions in water was 93 per cent of that measured in the presence of Triton X-100.The increase in hexokinase activity was irreversible even after very short exposure (90 s) to hypo-osmotic conditions. Total adenylate kinase activity was not affected by osmotic conditions. Adenylate kinase activity increased hyperbolically in supernatants prepared from mitochondrial suspensions with decreasing osmolarity of the sucrose or salt solutions. Besides monitoring adenylate kinase leakage as a measure of outer mitochondrial membrane disruption, mitochondrial swelling was followed by measurement of the turbidity of mitochondrial suspensions at 520 nm. The data has been interpreted in terms of binding of some hexokinase to the inner mitochondrial membrane.

    Notes: Neurotrophic ulceration of the face is a rare but well recognized sequel to division of the trigeminal nerve. 1,2 Trauma is an important contributory factor and thought to be due in part to paraesthesiae, which encourages picking and scratching, with resultant chronic and persistent ulceration. 3 A case is described of an 82-year-old woman with severe trigeminal neurotrophic ulceration which improved substantially with pimozide, given for treatment of unrelated paranoid symptoms. The possible relevance of this to the established use of pimozide in delusional parasitosis is briefly discussed.
