    Notizen: The problem of barrier leakage in surface pressure-surface area measurements has been overcome with a trough wherein the sample is compressed inside a band of teflon-covered spring steel strip. A computer-controlled stepping motor drive applies force to one side of the band, deforming it from a rounded triangle to a "v'' shape. The compression range extends up to 4:1 and is repeatable to ±2%. Calibration is conveniently and accurately done with a computer-interfaced video imaging and analysis system. The design greatly simplifies the construction of surface troughs and completely obviates the possibility of leakage at high surface pressures. The trough is useful for investigations involving high surface pressures and/or requiring long times.

    Notizen: Acetaldehyde and ethanol are usually thought to be produced in plant tissues as a mechanism to tolerate hypoxic conditions. We have found acetaldehyde and ethanol to be common in the vascular cambium and in the transpiration stream of trees. In nonflooded trees, acetaldehyde and ethanol concentrations averaged 130 and 40 μM in the cambium and 130 and 50 μM in the xylem sap, respectively. Ethanol concentrations in the transpiration stream and the cambium increased to as much as 5 mM upon flooding. Ethanol concentrations in the vascular cambium of Populus deltoides could not be eliminated by placing logs from nonflooded trees in a pure oxygen environment for as long as 96 h, but increased by almost 3 orders of magnitude when logs were exposed to low external partial pressures of O2. These results suggest that the vascular cambium was not hypoxic, despite the presence of acetaldehyde, ethanol and the enzymes for their synthesis.

    Notizen: To better understand the solution structure of spectrin, the environments of its tryptophan residues have been examined by fluorescence spectroscopy. The spectra and the extent of quenching by several quenching agents have been determined for intact spectrin and its α and β subunits. The arsenal of quenchers used in the study represented both hydrophilic and hydrophobic species including anionic, cationic and neutral compounds. Effects on spectrin fluorescence of ethanol and ionic strength, which extend and/or rigidify spectrin, and of glycerol, which is commonly used in electron microscopy of the protein, have also been assessed in the presence and absence of quenchers. Most of the tryptophans of spectrin are either internally quenched or are sequestered, hindering the approach of hydrophilic quenching agents. Both the spectral shape and the extent of quenching by acrylamide indicate that some tryptophans of the β subunit are slightly more exposed in the isolated chain than in the dimer. Similar effects on spectra and on quenching of the intact dimer and of the isolated β chain are seen when the ionic strength is reduced. Ethanol and glycerol reduce spectrin tryptophan accessibility to 2-p-toluidinyl napthalene-6-sulfonic acid (TNS). It therefore appears that low ionic strength, α-β association and neutral solute (or lowered dielectric constant) all induce a similar, but modest conformational change in the domain structure. The extent of TNS binding is not increased by lowering the ionic strength, suggesting that the expansion and/or stiffening of the molecule in low electrolyte solutions does not involve exposure of significant numbers of hydrophobic sites. © 1994 Wiley-Liss, Inc.
