    Notizen: A 32-year-old man, with a 12-year history of recurrent edema of the upper and lower extremities, was admitted in February 1998 due to edema of the right forearm and tiredness of 1-month duration. Examination revealed an ill-defined, itchy, slightly erythematous, indurated patch with excoriation on the right forearm (〈link href="#f1"〉Fig. 1). There was no history of drug intake, atopy, or asthma. He recalled, however, having experienced cold-induced Raynaud's phenomenon and intermittent arthralgia. In laboratory tests, there was a white cell count of 11.77 × 103/μL with 48% eosinophils (4830/μL). Liver function tests showed elevated serum glutamic pyruvic transaminase (SGPT) and alkaline phosphatase. Rheumatoid factor was positive. Total immunoglobulin E (IgE) concentration was elevated to 4703 U/mL (normal, 〈 378 U/mL), while other serum immunoglobulins, including IgG, IgA, and IgM, were normal. Bone marrow aspirate and biopsy showed normocellular marrow with an excess of mature eosinophils, but no evidence of blast cells. There was no evidence of parasitic infestation, and allergy skin test was negative. An extensive systemic evaluation showed no other organ involvement. A skin biopsy specimen revealed many eosinophils infiltrating the perivascular, periappendageal, and subcutaneous fat area and also within the vessel walls. Multiple microvascular thrombi with eosinophils were observed in the mid and lower dermis and subcutaneous fat (〈link href="#f2"〉Fig. 2). The eosinophils showed positive staining with eosinophil cationic protein (ECP) using the monoclonal antibody EG2. On the basis of the above findings, we diagnosed hypereosinophilic syndrome (HES). Treatment with 60 mg prednisolone daily was started and, within 7 days, his edema and itching sensation started to resolve. Follow-up laboratory and biopsy findings after 1 month returned to normal.     Notizen: Nano-sized titanium dioxides are incorporated into the interlayer spaces of ion-exchangeable layered perovskites, H1-xCa2-xLaxNb3O10 (x=0.00̃.75), by replacing the interlayer protons with positively charged TiO2 nano-sol particles or basic titanium glycolate complex (titanatrane). Powder X-ray diffraction analysis, UV-Vis absorption spectroscopy, thermogravimetric analysis, and N2 adsorption-desorption isotherm measurements show that quantum sized TiO2 particles are stabilized inbetween perovskite lattices to form micropores (SBET= 371̃10 m2/g). X-ray absorption spectroscopy at the Ti K-edge was used for investigating the local environment around Ti atoms constituting the interlayer pillars. According to the XANES spectra, the 'as-pillared' Ti species have the same local environments with those of precursory species, which subsequently converted into TiO2 clusters with rutile and anatase-like local structures when TiO2 nano-sol particles and titanatrane are used as pillaring species, respectively. It is also found that the local environment of TiO2 remains almost constant irrespective of the layer charge density, while the TiO2 pillar content and the microporosity increase with the latter.

    Notizen: This study was designed to investigate the influence of demineralized bone particles(DBP)/PLGA hybrid scaffold on angiogenesis and osteogenesis in a calvarial defect model.DBP/PLGA scaffolds were manufactured by solvent casting/salt leaching method, and each scaffoldcontained 0, 10, 20, 40, and 80 wt% DBP of PLGA, respectively. A total of 34 rats were operatedand bicortical holes were placed on their calvaria. The defects were filled with different ratioDBP/PLGA scaffolds. After 3, 7, 14, and 28 days, specimens were taken and, histologic,immunohistologic and RT-PCR analyses were carried out concerning number of vessels and densityof regenerated bone, and angiogenic activation. On days 7, in all experimental groups, boneformation occurred in a direction from defected margin of calvarium to center of implanted scaffoldand new vessel formation took place in front of the osteogenic regeneration front. We found that the20 and 40 wt% DBP/PLGA scaffold was superior in its ability to regenerate new bone, inducedmore intensive formation of microvasculature and expressed in a higher level of osteocalcin mRNAthan other groups

    Notizen: Recently, it has been studied tissue engineered technique as novel approaches fortreatment of the degenerative intervertebral disc (IVD). We designed the hybrid type of IVDmimicked scaffolds with poly(lactide-co-glycolide) (PLGA) and methoxypoly(ethyleneglycol)-poly([removed info]-caprolactone) (MPEG-PCL) diblock copolymers in order to application for the tissueengineered IVD. The MPEG-PCL solutions formed a gel-to-sol phase transitions as the temperaturewas increasesd. MPEG-PCL diblock copolymers were prepared by ring opening polymerization,and then nucleus pulposus (NP) cell was impregnated. Also, in order to restore annulus fibrosus(AF), we fabricated PLGA scaffold by solvent casting/salt leaching method. We confirmed disc cellfunction in manufactured scaffold through MTT assay in vitro and gross morphology and specialstaining in vivo for the possibility of the application of tissue engineering techniques

    Notizen: In order to application for the tissue engineered intervertebral disc (IVD), we designedthe synthetic/natural hybrid scaffolds with poly(lactide-co-glycolide) (PLGA) and small intestinesubmucosa (SIS). SIS has been widely used as a biomaterial because SIS consists of variouscollagens and cytokines. SIS, however, possesses disadvantages such as their weak mechanicalproperties and uncontrolled degradation. Novel composite scaffolds of PLGA/SIS weremanufactured by simple immersion method of PLGA scaffolds in SIS solution under vacuum. ThenSIS was crosslinked. Also, PLGA scaffolds and SIS sponges were manufactured by solventcasting/salt leaching and freeze-dried methods, respectively. We evaluated pore structure, porosity,water absorption ability and cell viability of three types of scaffolds for the application of IVD

    Notizen: In this study, the dipole switching and non-volatile memory functionality ofpoly(vinylidene fluoride-trifluoroethylene) (PVDF/TrFE)(72/28 mol%) random copolymer ultrathinfilms were analyzed. PVDF/TrFE(72/28) used as ferroelectric insulator in varying memory devicearchitectures such as metal-ferroelectric polymer-metal (MFM), MF-insulator-semiconductor(MFIS), MIS and ferroelectric field-effect transistors (FeFET) were examined using differentelectrical measurements. A maximum data writing speed of 1.69 MHz was calculated from theswitching time measured using MFM architecture. Compared to MFM, MFIS device architecture wasfound to be more suitable for distinguishing the ‘0’ and ‘1’ state using the capacitance-voltagemeasurement. With FeFET, the measured drain current (Id) as well as its memory window increasedwith decreasing channel length, thereby enabling the easier identification of ‘0’ and ‘1’ statecomparable to the MFIS case. The data obtained from this study will be useful in the fabrication ofnon-volatile random access memory (NVRAM) devices operating at lower voltage with faster dataR/W/E speed and memory retention capability

    Notizen: Polyvinylidene fluoride (PVDF) based electrospun nanoweb fibers with outstandingpiezo-, pyro- and ferroelectric behavior are being intensely studied by many researchers, especiallyfor touch-sensor applications. In order to further improve the advantageous characteristics of PVDFnanoweb fibers, we focused our attention on studying the effect of filling PVDF solution withcalculated amount of calcium chloride (CaCl2) or multi-walled carbon nanotube (MWCNT), andtheir electrospun nanoweb fibers were analyzed for the changes in β-crystalline phase, and itsassociated piezoelectric characteristics using a custom-made sensor set-up developed in our lab.FT-IR spectroscopy was used to confirm the changes in the β-crystalline content with varying contentof CaCl2 and MWCNT. SEM data revealed the reducing fiber diameter with increasing CaCl2 content.PVDF nanoweb subjected to pressure showed changes in touch sensing property as analyzed using anoscilloscope integrated with Labview program. Overall, the PVDF nanoweb containing the additivesused in our study exhibited greater sensitivity-in-touch for use in smart apparel applicationscompared to unmodified PVDF nanoweb, and the results are reported in detail here

    Notizen: Abstract Over-expression of Proteus vulgaris K80 lipase gene in Escherichia coli BL21 (DE3)/pKLE was achieved, with the enzyme being produced in an as active soluble form, using the T7 RNA polymerase system in a modified M9 salt or M9ZB medium. d-Lactose (55 mM) was used to induce gene expression and gave twice the lipase activity achieved with 0.4 mM IPTG. The expression of the lipase gene depended on the feeding rate of glucose being optimal at 12 g l−1 h−1.

    Notizen: Abstract To measure the quantity of tyrosine phosphorylated proteins in cells, we have used a flow cytometry technique with fluorescein isothiocyanate-labeled anti-phosphotyrosine antibody (FITC-αPY mAb). The analysis was applied to the phosphotyrosine titration and showed an optimum amount of FITC-αPY mAb (30μg/1×10cells). The staining specificity of our assay was tested by the addition of exogenous competitors, showing a specific inhibition by phosphotyrosine but not by phosphoserine, phosphothreonine, or tyrosine. The assay was also able to elucidate the inhibitory effect on tyrosine phosphorylation of genistein, a protein tyrosine kinase inhibitor. These results imply that immunofluorescent quantification assay using a flow cytometer could be a useful technique to determine the intracellular level of tyrosine phosphorylated protein.
