    Notes: Summary Isolated pancreatic islets of normoglycemic sand rats do not respond to 2.5 mM glucose with an enhanced glucagon secretion, which could be observed in normal Wistar rats. Arginine stimulates glucagon release in the presence of 2.5 mM glucose in Wistar rats as well as in sand rats. The secretion pattern is not caused by insulin deficiency since sand rat islets are characterized by an increased insulin secretion rate in vitro. This paradoxical glucagon secretion is not caused by a changed glucagon content but might be related to this species which is able to develop a diabetic syndrome spontaneously.

    Notes: Summary Two monoclonal Beta-cell surface antibodies M10H6 und K14D10 were obtained by fusion of spleen cells of Balb/c mice with the myeloma cell line P30. The monoclonal antibody M10H6 was induced by immunization with rat insulinoma cells finally boostered with disintegrated rat islets, whereas the K14D10 was generated after immunization with porcine proinsulin. Both monoclonals belong to the IgG2A isotype and were screened with insulin-producing rat insulinoma cells by an indirect immunofluorescence test as well as by a cellular enzyme linked immunosorbent assay. In addition to the cell surface binding on living Beta cells the monoclonals react with islets on cryostat sections of rat pancreas. The anti-islet cytotoxic potential of these monoclonals was measured by 51Chromium-release in the presence of complement or Fc-receptor bearing leucocytes using 51Chromium-labelled rat islet cells as target. Both antibody secreting hybridomas were propagated in syngeneic mice resulting in high levels of islet cell surface antibodies in ascites and sera from the recipient. High anti-islet cytotoxicity was mediated by ascites fluid, but no mouse developed hyperglycaemia. Furthermore, the repeated injections of the monoclonals into rats did not exert a diabetogenic action and failed to reduce the pancreatic insulin content although the attraction of the K14D10 to the pancreatic islets in vivo could be demonstrated. We conclude that islet cell surface antibody-mediated Beta-cell lysis in vitro may not be relevant to Beta-cell destruction in vivo.

    Description / Table of Contents: Summary Several investigations suggest changes of cellular metabolism and perhaps a precocious hemolysis of red cells caused by their prolonged stagnation in the spleen. Comparisons of red cell enzyme and substrate activity in splenic arterial and venous blood have been done in rabbits with experimentally induced stenosis of the splenic vein and in normal animals. In controls no difference of the parameters examined were found. In rabbits with splenic venous stenosis and retarded blood flow in the spleen the erythrocytes in the splenic vein showed significantly reduced values of G-6-PDH and GSSG-reductase. The results suggest “ageing” processes of red cells during their stagnation in the spleen. Besides, a part of the erythrocytes seems to be destructed precociously, because in all animals with splenic venous stenosis anemia and reticulocytosis have been observed.
    Notes: Zusammenfassung Verschiedene Untersuchungen legten den Verdacht nahe, daß die Stagnation der Erythrocyten in der Milz zu Änderungen des Zellstoffwechsels und damit vielleicht zu einer vorzeitigen Hämolyse führen können. Durch vergleichende Untersuchungen einiger Ferment-und Substrat-Aktivitäten der Erythrocyten in der Milzvene und Milzarterie wurde versucht, dieser Frage experimentell näherzukommen. Sie wurden an normalen Kaninchen und an Tieren mit künstlich gesetzter Milzvenenstenose durchgeführt. Bei normalen Tieren bestehen keine Unterschiede der gemessenen Größen. Bei den Tieren mit Milzvenenstenose, bei denen damit eine verlangsamte Blutströmung durch die Milz entsteht, wurden in den Erythrocyten der Milzvene signifikant erniedrigte Werte der G-6-PDH und der GSSG-Reductase gegenüber denen in der Milzarterie gefunden. Diese Befunde legen den Verdacht nahe, daß die Erythrocyten während der Stagnation in der Milz „älter“ werden. Ein Teil geht wohl auch vorzeitig zugrunde, denn es tritt bei allen Tieren mit Milzvenenstenose eine Anämie und eine Reticulocytose auf.

    Notes: Summary The nicotinamide antagonist 6-aminonicotinamide (6-AN) administered to rats prior to the second postnatal week drastically affected their motoric activity by producing consecutive reversible hypo- and hypermotoric stages. To obtain information on the validity of the putative link of correlated changes in brain RNA synthesis and motoric activity, neuronal and nonastrocytic glial cerebral and cerebellar in vitro RNA synthesis was determined during the hypo- and hypermotoric stages induced by in vivo administration of 6-AN given on the 7th day of a rat's life. Significant changes were apparent in the rate of neuronal cerebral and cerebellar RNA synthesis coinciding with the different phases of motor activity. However, only the changes in neuronal cerebral RNA synthesis were directly proportional to the changing pattern of motor activity. Measurement of the total number of RNA initiation sites present in neuronal cerebral chromatin, which was isolated during the phase of reduced and elevated muscular activity, revealed that the drug-induced changes in neuronal cerebral RNA synthesis could be directly attributed to a genetically repressed or activated state of the corresponding neuronal chromatin fraction. Nevertheless, although there was evidence suggesting a connection between changes in neuronal cerebral RNA synthesis and corresponding changes in muscular activity it was difficult to deduce any direct relationship between parameters owing to the absence of a convincing biological assay for assessing the role of RNA in motor function.

    Notes: Summary To determine the local gastrointestinal absorption of a new synthetic somatostatin analogue (SMS 201-995 = Sandostatin), an intestinal tube was passed in eight healthy volunteers and on different days an aqueous solution was administered at four different locations: stomach, proximal duodenum, ligament of Treitz and jejunum. In a follow-up study, an oro-ileal tube was passed in six of the original volunteers and the drug solution was administered in to the terminal ileum. The aqueous solution of SMS was rapidly absorbed from the gastrointestinal tract after local application, and it was well tolerated. Absorption of the drug from the different sites was comparable, although there was a tendency to decreased peptide absorption after ileal administration. Absorption of the drug was quite variable between the subjects and the different locations. The dose-corrected systemic availability relative to subcutaneous administration in another study was 0.28%. However, significant plasma SMS concentrations were achieved, suggesting that oral delivery of the polypeptide may eventually be possible for long-term treatment of a variety of disorders.

    Notes: Abstract Cells of Bacillus subtilis 168+ were labeled with 32P-orthophosphate during the process of sporulation, germination and outgrowth. By two-dimensional gel electrophoresis, at least 30 protein species were found to be radioactively labeled; 30% of these were modified by phosphorylation. Significant changes in the protein phosphorylation pattern during growth and cellular differentiation could be demonstrated. Using γ-32P-ATP evidence for an ATP-dependent protein kinase was also obtained. Under these conditions 4 proteins with a molecular mass of 109 600; 103 100; 73 300 and 32 200 Da were found to be phosphorylated.

    Notes: Summary The aim of the present study was to evaluate the diagnostic accuracy of serology by using new assays for the detection of genus and species-specific IgG, IgM, IgA and secretory IgA antibody in female sex workers. Cervical swabs and first void urine (FVU) from 314 female sex workers were submitted to nucleic acid amplification by ligase chain reaction (LCx, Abbott). Concomittantly, blood samples were tested for the presence of IgG, IgM and IgA antibodies using a genus-specific assay (rELISA, Medac) and species-specific test (SeroCT, Orgenics). Chlamydia trachomatis infection was detected in a total of 30 (9.6%) female sex workers by LCR. With rELISA, seroprevalences for IgG, IgM and IgA antibody to Chlamydia were 88.9%, 19.1% and 62.7%, respectively. IgG and IgA antibody prevalences against C. trachomatis (SeroCT) were 65.0% and 23.9%, respectively. In comparison to the positive LCR results obtained from cervical swab and/or FVU, the sensitivity of rELISA for Chlamydia IgG, IgA and IgM detection was 93.9%, 83.3% and 16.7%, respectively. With SeroCT, the sensitivity for C. trachomatis-specific IgG and IgA detection was 86.7% and 33.3%, respectively. The specificities of both serologic tests in comparison to LCR were very slow. C. trachomatis infection of the lower genital tract is very low. According to our results, serologic testing for Chlamydia can exclude active infection of the lower genital tract with a high reliability (≥ 95%). However, detection of C. trachomatis can only be reliably achieved by nucleic acid amplification assays.

    Description / Table of Contents: Zusammenfassung Elektrisch gereizte Vorhofpräparate des Meerschweinchens wurden in einem Tyrodebad suspendiert, dessen pH durch Begasung mit verschiedenen CO2-Konzentrationen auf 7,0, 7,4 bzw. 7,8 eingestellt wurde. Kontraktionskraft und maximale Kontraktionsgeschwindigkeit der Vorhöfe wurden durch diese pH-Änderungen nicht beeinflußt. Der positiv inotrope Effekt von Tyramin, Isoproterenol und Calcium war bei pH 7,0 abgeschwächt, bei pH 7,8 verstärkt. An narkotisierten Katzen wurde durch Beatmung mit 10 bzw. 20% CO2 eine respiratorische Acidose mit pH-Werten von 7,15 bzw. 7,0 erzeugt. Dabei traten eine mäßige Zunahme der Herzfrequenz, der maximalen Druckanstiegsgeschwindigkeit (dp/dtmax) und des systolischen Femoralisdruckes ein. Die Wirkung aller untersuchten Pharmaka war in Acidose abgeschwächt, wobei sich aber folgende quantitative und qualitative Unterschiede ergaben: 1. Die blutdrucksenkende Wirkung des Isoproterenols und die blutdrucksteigernde Wirkung des Noradrenalins waren stärker abgeschwächt als ihre positiv inotrope Wirkung am Herzen. 2. Die pressorische Wirkung des Angiotensins und Noradrenalins war weniger beeinträchtigt als die des Naphazolins. 3. Die blutdrucksenkende Wirkung des Acetylcholins war im Gegensatz zu der des Isoproterenols weniger abgeschwächt.
    Notes: Summary Experiments were performed on guinea pigs isolated atria which were stimulated electrically. If the pH of the tyrode solution was changed by variation of pCO2 no alteration of force of contraction and maximal velocity of contraction (dT/dtmax.) occured. The inotropic effects of tyramine, isoproterenol and calcium were diminished at pH 7.0 and increased at pH 7.8. During respiratory acidosis (pH 7.0) anaesthetized cats showed a small increase in heart rate, left ventricular dp/dtmax. and arterial blood pressure. The effects of all tested drugs were diminished in acidosis, whereby the following qualitative and quantitative differences appeared: 1. The depressive effect of isoproterenol and the stimulating effect of norepinephrine on arterial blood pressure were much more diminished than the positive inotropic effect of these drugs. 2. The effect of angiotensin and norepinephrine on arterial blood pressure was less affected than that of naphazoline. 3. The effect of acetylcholine on arterial blood pressure was scarcely diminished in contrast to that of isoproterenol.
