    Notizen: Abstract The pericentromeric heterochromatin of meiotic trivalents formed by the Robertsonian (Rb) chromosomes and the two homologous acrocentrics in the house mouse was evaluated by static cytophotometry after selective staining. To reveal pericentromeric heterochromatin specifically, C-banding Giemsa and Hoechst 33258 stains were utilized. Five different Rb chromosomes were investigated and none of them possessed less pericentromeric heterochromatin than the sum of the two homologous acrocentrics. Moreover the total A-T (DAPI) and DNA (PI) content was quantitatively evaluated, by flow cytometry, in G0/G1 nuclei belonging to four different Rb mouse populations, karyotypically characterized by the presence of up to nine Rb chromosomes. Again there were no significant difference, of DAPI and PI content, in the Rb populations nor between any of them and the NMRI/HAN strain with forty aerocentric chromosomes. We conclude that the main consequence of Robertsonian processes (i.e. the rapid variation of the karyotype structure) does not imply detectable quantitative variation in the genome portion involved in the Rb process. We also discuss the possibility that the high rate of Rb exchange in the house mouse could be favoured by the simultaneous effects of undetectable losses of chromosomal material, high repetitiveness of the DNA involved, the presence of the same major type of satellite DNA over each chromosome and the all acrocentric constitution of the karyotype.

    Notizen: Summary Three familial cases of idiopathic hypertrophy of the masticatory muscles are reported. The muscular enlargement was noted in late childhood or early adult life, was bilateral, insidious, painless and non-progressive. Electrophysiological and laboratory findings were normal in one case. CT examination, performed in two cases, showed homogeneous bilateral enlargement of masseter muscle bulk consistent with true hypertrophy. Two patients also showed congenital malformations. A review of the literature showed that this disorder is very rare.

    Beschreibung / Inhaltsverzeichnis: Sommario Riportiamo i risultati del trattamento con plasmaferesi (PE) e immunoglobuline (IVIg) in 36 su 50 pazienti con sindrome di Guillain-Barré (GBS) reclutati attraverso uno studio di incidenza condotto in Emilia-Romagna. Il confronto fra PE e IVIg non ha evidenziato differenze significative per quanto concerne la capacità di migliorare il decorso della malattia. Complessivamente, nei trattati con IVIg e PE il 25% e l'11,1% rispettivamente erano guariti ad un mese, mentre il 58,3% e il 55,5% erano migliorati di almeno un grado. Questi risultati sono in accordo con le conclusioni del Dutch GBS Trial per ciò che concerne l'outcome a un mese. Nessuna ricaduta è stata osservata nei due gruppi. Inoltre, non è stata osservata nessuna differenza nell'outcome clinico a 1 e 3 mesi tra i pazienti che ricevettero una sola o due terapie. Un secondo ciclo di terapia non sembra migliorare significativamente il decorso clinico della malattia. PE e IVIg sono entrambe sicure ed efficaci nella terapia della GBS.
    Notizen: Abstract We report the effects of treatment with plasma-exchange (PE) and intravenous immune globulins (IVIg) in 36 out of 50 patients with Guillain-Barré syndrome (GBS) recruited by an incidence study in the Emilia-Romagna region of Italy. Comparison of the patients treated with PE and IVIg showed no significant differences in terms of effectiveness in improving the clinical course of GBS: at one month, respectively 11.1% and 25% had recovered, and 55.5% and 58.3% had improved by at least one grade. These results are in agreement with those of the Dutch GBS trial. No relapses were observed in either group. Moreover, our results showed no difference in clinical outcome at 1 and 3 months between the patients receiving only one therapy and those receiving two; a second cycle of therapy did not seem to improve the clinical course of the disease significantly. We conclude that PE and IVIg are both safe and effective therapies for GBS.

    Notizen: Summary C6 glioma cells (CCL 107) were cultured for three days and then treated withcis-dichlorodiamineplatinum (cis-DDP) at doses of 0.2–10 µg/ml medium. Changes in DNA synthesis and DNA content, as well as morphology of cells and chromatin distribution, were examined from the first post-treatment day onwards. The number of cells labelled with [3H]thymidine, detected autoradiographically, decreased after treatment with 0.2–10 µg/ml by approximately one half on post-treatment day 1 and diminished further by the third day after treatment. The labelled cells were entirely absent only after treatment with 10µg/ml, 7 days post-treatment. Mitoses decreased from 1.4–0.6% by post-treatment day 1 and completely disappeared by day 3 (1 µg/ml). Feulgen cytophotometry and propidium iodide cytofluorimetry revealed accumulation of cells in the S-phase, especially the latter part (0.5 and 1.0 µg/ml, post-treatment day 1) and subsequently also in G2 phase (post-treatment day 3). Incomplete cyto- and karyokinesis in some cycling cells was indicated by an increased number of binucleate cells and nuclei of higher ploidy classes. Labelled cells with intermediate DNA values were, on average, labelled less intensively, as was revealed by simultaneous measurements of DNA content and [3H]thymidine incorporation. Some cells displayed reduction in grain density over heterochromatin clumps. This would be in agreement with the late S-phase block of DNA replication. After post-treatment day 3 the density of cells in cultures was substantially lower. This was due to slowed transversing through the cell cycle and cell death occurring after post-treatment day 1 with higher doses or after day 2 with lower doses (up to 1 µg/ml). The size of the nuclei of surviving cells enlarged initially (post-treatment day 1) and later (day 7) giant cells with long, branched fibres similar to those of reactive astrocytes occurred. Texture analysis of Feulgen-stained nuclei revealed that the chromatin of cells treated withcis-DDP became less evenly distributed. This might be due either to the direct influence ofcis-DDP on the DNA molecule, or mediated by changes in cytoskeleton and cAMP levels described earlier.
