    Notes: Summary To evaluate possible influences of dietary intervention on the composition of very low-density lipoproteins (VLDL), ten subjects with Type 2 (non-insulin dependent) diabetes mellitus received a hypocaloric regimen. Fifteen healthy subjects served as controls. Ultracentrifuged VLDL were analysed as cholesterol, triglycerides, apolipoprotein B (apo B), and the soluble apolipoproteins C and E (polyacrylamide gel electrophoresis in urea and densitometry) before the study, after 2 weeks and then after 3 months. Compared with the control subjects, the content of cholesterol and apo E in the VLDL was elevated in the diabetic subjects, while the area ratio of apo C-II to apo C-III1 was lowered. After diet the reduction in VLDL was accompanied by compositional changes: a decrease of the cholesterol/triglyceride ratio and of the apo E/apo C area ratio. The apo C-II/apo C-III1 area ratio remained unaffected. We conclude that one beneficial effect of therapeutic intervention in diabetes may lie in lowering the level of possibly atherogenic VLDL-components.

    Description / Table of Contents: Summary We compared the total lipids, the total esterified fatty acids, the Lipid phosphorous and the cholesterol fractions in isolated leukocytes of 14 chronic lymphatic leukemias, 10 chronic myeloic leukemias and 5 acute myeloblastic leukemias with the lipid fractions in isolated leukocytes of normal persons. The total lipids and the total esterified fatty acids gave a relation of 1:2:3 in chronic lymphatic leukemia: chronic myeloic leukemia: normal. If at all esterified cholesterol was present only in small amounts. These results are compared with the findings in literature.
    Notes: Zusammenfassung Bei 14 Patienten mit chronischer lymphatischer Leukämie, 10 Patienten mit chronischer myeloischer Leukämie und 5 Patienten mit akuter Myeloblastenleukämie wurden in isolierten Leukocyten die Gesamtlipide, die veresterten Fettsäuren, der Lipidphosphor, das freie Cholesterin und das Gesamtcholesterin bestimmt und mit den bei Normalpersonen gefundenen Werten verglichen. Dabei fand sich bei Gesamtlipiden und veresterten Fettsäuren ein Verhältnis von 1:2:3 entsprechend chronische Lymphadenose: chronische Myelose: normal. Verestertes Cholesterin war — wenn überhaupt — nur in geringen Mengen nachweisbar. Die Ergebnisse werden mit den Angaben in der Literatur verglichen.

    Description / Table of Contents: Summary The phospholipid composition of leucocytes from 11 patients with chronic myeloic leukemia and 3 patients with chronic lymphatic leukemia were investigated by thin-layerchromatography. The results are compared with literature. The main phospholipid fractions are lecithine and colamincephaline. There are differences of phospholipid-composition between leukocytes and plasma on one hand and between lymphatic leukemia and myeloic leukemia/normal leukocytes on the other.
    Notes: Zusammenfassung Es wird über die dünnschichtchromatographische Bestimmung der einzelnen Phospholipoidfraktionen in isolierten Leukocyten von 11 Patienten mit chronischer Myelose und 3 Patienten mit chronischer Lymphadenose berichtet. Die Ergebnisse werden mit den in der Literatur mitgeteilten Werten verglichen. Lecithin und Colaminkephalin machen den Hauptanteil der Phospholipoide aus. Unterschiede bestehen einmal gegenüber den Plasmawerten und zum anderen zwischen Lymphadenosen einerseits und Myelosen und Normalen andererseits.

    Description / Table of Contents: Summary The activities of 8 key-enzymes (glucose-6-phosphate-dehydrogenase, 6-phosphogluconate-dehydrogenase, hexokinase, phosphofructokinase, pyruvate kinase, glycerol-3-phosphate-dehydrogenase, malate dehydrogenase, phosphoglucomutase) have been estimated in the subcutaneous adipose tissue of 27 patients with hyperlipemia of different origin. We found that the activities of glucose-6-phosphate-dehydrogenase and hexokinase were diminished significantly, what means a lowered utilisation of glucose. From these results in connection with a diminished activity of glycerol-3-phosphate-dehydrogenase it can be concluded that lipogenesis in human adipose tissue is diminished at the same time.
    Notes: Zusammenfassung Im subcutanen Fettgewebe von 27 Patienten mit Hyperlipidämien verschiedener Genese wurden die Aktivitäten von 8 Schlüsselenzymen bestimmt (Glucose-6-Phosphat-Dehydrogenase, 6-Phosphogluconat-Dehydrogenase, Hexokinase, Phosphofructokinase, Pyruvatkinase, Glycerin-3-Phosphat-Dehydrogenase, Malatdehydrogenase, Glucophosphomutase). Dabei fand sich eine hochsignifikante Herabsetzung der Glucose-6-Phosphat-Dehydrogenase- und der Hexokinase-Aktivität, woraus auf eine verminderte Glucoseverwertung geschlossen werden kann. In Verbindung damit spricht die deutlich erniedrigte Aktivität der Glycerin-3-Phosphat-Dehydrogenase für eine gleichzeitig herabgesetzte Lipogenese im menschlichen Fettgewebe.

    Description / Table of Contents: Summary This is a report on changes of concentration of 9 serum proteins and 4 lipid fractions in 8 patients treated with L-Asparaginase during a period between 14 and 36 days. Under therapy a decrease of initial concentration of albumin, α2-macroglobulin, coeruloplasmin, α1-antitrypsin, transferrin and α2-haptoglobin was observed, whereas after treatment the proteins showed a tendency of returning to the initial concentrations. Concerning the immunoglobulins, an inverse behaviour was found, i.e. there was an increase of IgG, IgA and IgM during Asparaginase therapy and a gradual decrease after therapy. With serum lipids the following changes were seen: the concentrations of the free fatty acids rose during asparaginase treatment and the total lipids, triglycerides and cholesterol levels diminished within the first 14 days of therapy. After treatment a normalizing tendency of the lipid fractions was observed among those patients examined. From these results it may be concluded that L-Asparaginase influences the protein and lipid metabolism even of normal cells.
    Notes: Zusammenfassung Es wird über Konzentrationsänderungen von 9 Serumproteinen und 4 -lipidfraktionen bei 8 Patienten mit malignen Tumoren und Hämoblastosen berichtet, die mit L-Asparaginase über einen Zeitraum zwischen 14 und 36 Tagen behandelt wurden. Unter Asparaginase-Therapie konnte bei nahezu allen Patienten ein Absinken der Ausgangskonzentration von Albumin, α2-Makroglobulin, Coeruloplasmin, α1-Antitrypsin, Transferrin und α2-Haptoglobin beobachtet werden, während diese Proteine nach Absetzen der Therapie eine Tendenz zur Rückkehr zu den Ausgangskonzentrationen erkennen ließen. Für die Immunglobuline konnte ein gegenläufiges Verhalten gefunden werden, d. h. Zunahme von IgG, IgA und IgM unter Asparaginase-Therapie und eine allmähliche Abnahme nach Absetzen der Therapie. Bei den Serumlipiden fanden sich folgende Veränderungen: die freien Fettsäuren stiegen während der Behandlung an, die Gesamtlipide, Triglyceride und das Cholesterin fielen innerhalb der ersten 14 Behandlungstage ab. Nach Absetzen der Therapie zeigte sich bei den nachuntersuchten Fällen eine Normalisierungstendenz. Aus den Befunden wird der Schluß gezogen, daß die L-Asparaginase den Protein- und Lipidstoffwechsel auch von normalen Zellen beeinflußt.

The enzymes were selected in regard to possible changes in different metabolic diseases. The results are compared to the literature and the differences are discussed.

Die Ergebnisse werden mit den Angaben in der Literatur verglichen und die vorhandenen Unterschiede diskutiert.

    Description / Table of Contents: Zusammenfassung Im subcutanen Fettgewebe von Mensch und Schwein sowie im epididymalen Fettgewebe der Ratte wurden Enzymmuster bestimmt: Glucose-6-Phosphatdehydrogenase (EC 1.1.1.49), 6-Phosphogluconatdehydrogenase (EC 1.1.1.44), Hexokinase (EC 2.7.1.1), Phosphofructokinase (EC 2.7.1.11), Pyruvatkinase (EC 2.7.1.40), Glucophosphomutase (EC 2.7.5.1), Glycerin-3-Phosphatdehydrogenase (EC 1.1.1.8), Malatdehydrogenase (EC 1.1.1.37), Glutamatpyruvattransaminase (EC 2.6.1.2), Glutamatoxalacetattransaminase (EC 2.6.1.1), Glutamatdehydrogenase (EC 1.4.1.3), Citrate Cleavage Enzyme (EC 4.1.3.8), Malic Enzyme (EC 1.1.1.38), Glycerokinase (EC 2.7.1.30), Glucose-6-Phosphatase (EC 3.1.3.9) und Fructose-1,6-Diphosphatase (EC 3.1.3.11). Trotz der unterschiedlichen Lokalisation der Gewebe und des verschiedenen Gehaltes an löslichem Extraktprotein (Ratte:Mensch:Schwein=21,7±1,6:21,0±1,9:5,4±0,5mg/g Frischgewicht) finden sich annähernd übereinstimmende Enzymaktivitätsmuster. Bei allen drei Species ist die Phosphofructokinase das limitierende Glykolyse-Enzym und erfolgt der Glucoseabbau offensichtlich überwiegend über den Pentose-Phosphat-Shunt; eine weitere Gemeinsamkeit ist das Fehlen von Glycerokinase-Aktivität. Bezogen auf Extraktprotein liegen jedoch fast alle Enzymaktivitäten im menschlichen Fettgewebe um annähernd eine Zehnerpotenz niedriger als bei Ratte und Schwein. Im Gegensatz zu diesen beiden Species ist im Humanfettgewebe kein Citrate-Cleavage-Enzyme nachweisbar.

The conduct of soluble extract-protein was equal in rat (21.7±1.6 mg/g) and man (21.0±1.9 mg/g) but different in pig (5.4±0.5 mg/g). In all three species the pattern was nearly the same; phosphofructokinase was the rate-limiting enzyme of glycolysis; most of the glucose degradation in white adipose tissue seems to be done via the pentose-phosphate-shunt; there was no measurable activity of glycerokinase. In human adipose tissue—where nearly all enzyme activities were about ten times lower—there was no citrate cleavage enzyme either.
