    Notes: The arterioles and capillaries supplying the lymphatic nodules and marginal zone (in part) in the rhesus monkey's spleen arise from a follicular artery branch of the central artery. This follicular artery first undergoes a series of convolutions and branchings, out of which a parallel array of tightly bound arterioles and capillaries, an anteriorlar capillary bundle (ACB), leading into the central part of the nodule is formed. From the proximal end, side, and distal end of the ACB, arterioles and capillaries radiate outward through the mantle zone of the nodule to terminate in the marginal zone. Some capillaries are formed within the bundle and others not until an arteriole nearly reaches the marginal zone.A similar ACB and capillary distribution is found in the human spleen. However, the supplying arteries, for the most part, come from recurrent penicillar arteries which pass close to the central artery before forming the bundle. This artery does not undergo the convolutions found in the rhesus monkey. In the monkey, the bundle contains a higher proportion of arteriolar vessels. It appears that in human and rhesus monkey spleens, the ACB replaces the internal capillary net of Jäger (′29).

    Notes: A marginal sinus, formed by the anastomosed terminations of white pulp capillaries, bounds the inner edge of the marginal zone. The endothelial lining on the nodular surface is complete. Pores appear along the lateral aspect thus allowing blood to pass freely into the interstices of the marginal zone. A dense aggregation of Marshall's metalophil cells occurs in the region of the marginal sinus. Such cells are lacking in the marginal zone.Intravenous injections of particulate matter were used to determine the course of blood flow and phagocytic potential. Ten minutes after injections, particles were found to be dispersed among the cells of the marginal zone. None were found in the marginal metalophils. In four hours the particles had cleared the marginal zone and were within red pulp macrophages. In a few rats, the marginal metalophils reacted positively to tests for iron. It is concluded that marginal metalophils represent a large reserve of potential phagocytes situated at a point where blood first leaves capillaries to enter tissue interstices.

    Notes: Questions concerning the nature of the intermediary vascular pattern make up a considerable portion of the literature concerning splenic morphology. Evidence indicates there is a special association between the reticuloendothelial system (RES) and the intermediary capillary terminations. This study was undertaken to characterize the distribution of the RES in the rabbit spleen and to correlate this with the vascular pattern described by Snook ('58). Like cells of the RES throughout the animal body, splenic RE cells can be demonstrated by several histochemical and functional methods. They are metalophilic (argyrophilic), possess several hydrolytic enzymes, and phagocytose injected particulate matter. Cells demonstrated by these methods are found in the marginal zone, and in the lumen of the red pulp sinuses but most are localized in the red pulp cords. This distribution corresponds closely to the termination of arterial capillaries in the marginal zone and pulp cords in the rabbit spleen and supports the general hypothesis that there is a unique and intimate relationship between the vascular system and the RES at these points.The sinus lining cells react characteristically to the techniques employed. They are sporadically metalophilic, strongly positive for nonspecific esterase (endoplasmic reticulum), negative for acid phosphatase (lysosomal), and occasionally contain small particles of injected matter. They probably represent a functionally distinct cell population.

    Notes: The major arteries which supply the follicular capillaries in the human spleen do not arise as they do in most mammals as lateral or radial branches from the central artery but come from penicillar arteries which penetrate the marginal zone and enter the follicle at various points around its circumference. Such arteries may have a very short course through the red pulp or they may pursue very long courses.Upon entering the follicle, these arteries branch a number of times, the branches remaining together in a tight array of parallel arterioles along with capillaries formed from them, the whole bundle being enveloped by a reticular fiber sheath. There is thus formed an arteriolar-capillary bundle. The whole bundle may branch. From the sides, especially from its central end, arterioles and capillaries radiate out to all parts of the follicle to terminate in the marginal zone or in the follicle itself.

    Notes: The effects of the estrogenic compounds, diethylstilbestrol (DS) and diethylstilbestrol diphosphate (DSP) on morphologically demonstrable characteristics of reticuloendothelial (RE) cells were studied in rat spleen. These included metalophilia, acid phosphatase, nonspecific esterase and phagocytosis. Routine histological and histochemical techniques were used to determine the distribution of metalophilic cells, hydrolytic enzymes, the presence of intracytoplasmic PAS-positive material, hemosiderin sequestration, and the ability to phagocytose injected carbon particles and bacterial organisms.Changes in these parameters associated with increased RE function were observed in several regions of spleens of DS or DSP-treated animals. The marginal metalophils at the periphery of the nodule, normally not phagocytic in animals of the age studied, became phagocytic and demonstrated increased metalophilic and enzymatic activity. The red pulp was characterized by increased densities of cells in pulp cords demonstrating metalophilia, hydrolytic enzyme activity, PAS positivity and hemosiderin. Phagocytosed material was limited to cells in the pulp cord. The reactions of the sinus-lining cells were not affected by this treatment. Results indicated that the marginal metalophils have the capacity to become fully functional RE cells under the influence of these drugs.
