    Notizen: Abstract The applicability of a chlorophyll fluorescence assay for kanamycin (Km) resistance screening in transgenic tobacco (Nicotiana tabacum) and Arabidopsis thaliana plants was investigated. In wild-type leaves incubated in the presence of 200 mg/l Km, a decrease in maximum variable fluorescence ((Fv)m) and a significant increase in constant fluorescence (Fo) were observed. Using (Fv)m/Fo as a screening parameter, we were able to distinguish Km-treated samples from untreated samples within 4 days. This parameter was applied to Km resistance screening using tobacco plants transformed with the nptII gene via Agrobacterium. Among 74 shoots selected on medium containing 200 mg/l Km, 37 plants were scored as Km sensitive by the chlorophyll fluorescence assay. These 74 scorings proved to be accurate, as reconfirmed by (1) polymerase chain reaction amplification of the transgene, (2) enzymatic assay of neomycin phosphotransferase and (3) leaf disc assay. Using the chlorophyll fluorescence assay, we could also screen 3-week old Arabidopsis plants carrying the nptII gene. These results clearly demonstrate the reliability and efficiency of this nondestructive assay for Km resistance screening of transgenic plants.

    Notizen: Continuous vinyl acetate emulsion polymerization was carried out by using a mixed surfactant system of “Aerosol” OT (AOT) and sodium dodecyl sulfate (SDS). It is well known that the AOT reduces surface tension considerably and has stronger monomer solubilization power than SDS. In this work, particular attention is given to observe the effects of the mixed surfactant on the molecular weights and polydispersities. In comparison with experiments using only SDS, the particle size appears similar, in the range of 190-280 nm. The conversion increased substantially to the 70% range from the 50% level observed with only SDS. All experiments were conducted at 60°C with a mean residence time of 30 min. The total surfactant concentration was maintained at 0.03 mol/L water for all experiments. It was found that once some AOT was included in the surfactant, the behavior was very similar to that observed with only AOT. The number average molecular weights were found to decrease substantially while the polydispersity increases dramatically to about 12. It is believed that AOT may act as a significant chain transfer agent in the polymerization.

    Notizen: Molecular structures for cured, thermoset resins have been examined by analyses of resin extracts by gel permeation chromatography. Numerical interpretation of leached oligomeric fractions coupled with kinetic reaction theory yields microscopic estimates of the network's structure, including crosslink average molecular weight. Leached monomeric fractions describe extent of cure. Analyses of test data incorporate irreversible, mechanical deformations for neat resin castings and for filament wound, Kevlar 49 composites. Heat distortion temperatures correlate with crosslink architecture. Short beam shear strength data for naval ordinance laboratory rings and pressure vessel burst performance are examined in terms of the resin's molecular structure. Variations in resin cure observed by positive feedback via chromatography indicate fluctuations in extent of crosslink development. Test specimen performance correlates with these microscopic, molecular distributions. Resin applications include filament wound composites for the aerospace industry, anhydride cured epoxies, and amine cured epoxies. The former resin is an elastic body at ambient test conditions. A third resin, a crosslinked resin of 1,2 polybutadiene/t-butylstyrene, is in final developmental stages and may exhibit a higher degree of viscoelastic behavior.

    Notizen: A study of the temperature dependence of Mooney scorch time was carried out by using two grades of natural rubber, Hevea Brasiliensis(SMR L and SMR 10), and on synthetic rubber, styrene - butadiene rubber (SBR), in the temperature range of 100-180°C. Results show that the scorch time for SBR system is greater than that of the other grades of natural rubber. This is attributed to the lower degree of unsaturation in SBR. Effects of 2-mercaptobenzothiazole (MBT) and other types of accelerators on the scorch properties were also investigated and discussed. One black-filled compound was used to study the dependence of carbon black on the scorch property, and data indicate that the effect is more evident for temperature lower than 100°C.

    Notizen: The adhesive properties of a series of oligomers prepared from 2-isocyanatoethyl methacrylates (IEM) and/or m-iso-propenyl-α,α-dimethylbenzyl isocyanate (TMI) and various acrylates or methacrylates were studied. The bond strength of bone, dentin, or soft tissue specimens joined with these oligomers respectively to bone, dental composite restorative, or denture base resin were determined by tensile adhesion or shear tests. These oligomers are more effective in forming stronger bonds to bone than are other tissue adhesives. Fracture occurs cohesively, usually within the bone. Thermocycling in water for 1 week between 5°C and 55°C did not decrease adhesion indicating that exposure to water or thermal shock produced no deterioration of the bond. Tensile adhesion of bovine or human dentin joined to composite restorative resin by means of the oligomers is similar to that of the best dental bonding agents such as Gluma (glutaraldehyde and 2-hydroxyethyl methacrylate) or ferric oxalate + Nphenylglycine + dimethylacryloxyethyl-pyromellitate. These oligomers also strongly bond soft tissues and calfskin and to acrylic resins and composites.

    Notizen: A series of methacrylate oligomers containing pendant isocyanate groups were synthesized by reacting 2-isocyanatoethyl methacrylate (IEM) and/or m-isopropenyl- α, α-dimethylbenzyl isocyanate (TMI) in ethoxyethyl acetate with methacrylates ranging from methyl to stearyl methacrylate or allyl-, cyclohexyl-, glycidyl-, i-bornyl-, or dicyclopentenyloxyethyl methacrylate. The oligomers which are stable at room temperature were characterized by IR for NCO, ester, and C = C groups and by their refractive indices. They have a small number of residual double bonds and a molecular weight low enough so that the compounds are liquids at room temperature and dissolve readily in esters and chlorinated hydrocarbons. HPLC showed no residual monomer. GPC and intrinsic viscosity of selected oligomers indicated a molecular weight range from 1400 to 2600. Isocyanate groups were determined titrimetrically and ranged from 15.9% to 5.1%. Concurrent studies have demonstrated that these oligomers bond strongly to hard and soft tissues. Thus, subject to their biocompatibility they could find many applications as tissue adhesives.

    Notizen: The retentions of 11 explosives on two commercially RP-18 and Si 100 stationary phases and one prepared 3-[9′-(10′-methylanthryl)]- propylsilane stationary phase (MAPS-phase) were examined by modified mobile phases. The evaluation of these three stationary phases results as follows: (1)Most explosives were effectively separated on a column of MAPS-phase not only with methanol/water (75/25-85/15) as mobile phase but also with n-hexane/dichloromethane (75/25) as mobile phase.(2)Few explosives were effectively separated on a column of RP-18 with methanol/water (75/25-85/15) as mobile phase.(3)The separation of most explosives on Si 100 was achieved only under normal phase condition such as n-hexane/dichloromethane (75/25) as mobile phase.
