    Notizen: Abstract A rhamnose-containing microbial polysaccharide has been produced by Klebsiella sp. I-714. strain. The polysaccharide has been used as a source for the obtention of L-rhamnose. A biotechnological process has been developed to purify the hydrolyzed polysaccharide (EPSH), which contains rhamnose, galactose and glucuronic acid. Microbial removal of galactose is followed by a continuous chromatographie separation of glucuronic acid to render pure rhamnose. The technical feasibility of the process has been studied, with special emphasis on microbial inhibitor's removal.

    Notizen: Abstract Fluidized-bed bioreactors (FBRs) with immobilized cells have been used successfully at Oak Ridge National Laboratory and at the Autonomous University of Barcelona for several applications, including high-productivity yields of ethanol. During the development of these processes, experimental results have revealed differences between three-phase fluidized biocatalyst systems and those systems typically used for hydrodynamic studies. These differences appear to be related to the physical properties (density, viscosity, surface tension) of the phases and, in particular, the low density of the solid particles. Parameters were calculated from these physical properties following the approach of Baker for two-phase flow. A map of these parameters was proposed previously and separated regions of satisfactory and unsatisfactory fluidization of a three-phase immobilized-yeast FBR fluidized with air. This same map was used for an FBR with immobilized Zymomonas mobilis that was liquid fluidized. The stability regions on the map corresponded to the two different experimental operating systems. This indicates the possible general applicability of this approach to biological FBRs.

    Notizen: The continuous production of a hybrid antibiotic by a transformed strain of Streptomyces lividans TK21 in a three-phase fluidized bed is studied. Cell aggregates, known as pellets, are used as immobilized cell particles in the bioreactor. A methodology to prepare pellets of a suitable size and morphology is developed. The continuous production of the antibiotic is studied on the basis of decoupling cell growth and antibiotic production, by means of phosphate limitation in the growth medium. The best results are achieved at D = 0.021 h1, with alternate feeding of 0 and 0.05 m M phosphate media. Continuous production of the antibiotic can be maintained at satisfactory levels for periods of 60 days, and stable operation of the bioreactor is achieved during 85 days. Finally, the evolution of the internal structure of the pellets during continuous fermentation is studied. © 1997 John Wiley & Sons, Inc. Biotechnol Bioeng 53: 601-610, 1997.

    Notizen: Abstract The present work describes the genetic modification of a hybridoma cell line with the aim to change its metabolic behaviour, particularly reducing the amounts of ammonia and lactate produced by the cells. The cellular excretion of ammonia was eliminated by transfection of a cloned glutamine synthetase gene. The metabolic characterisation of the transformed cell line includes the analysis of the changes introduced in its intracellular metabolic fluxes by means of a stoichiometric model. Furthermore, the reduction of lactate accumulation was attempted through an antisense mRNA approach, aiming to generate a rate limiting step in the glycolytic pathway, thus lowering the glucose consumption rate. The physiological results obtained with the transformed cells are discussed. A maximum reduction of about 47% in the glucose consumption rate was obtained for one of the transformations. However a main drawback was the lack of stability of the transformed cells

    Notizen: Summary A novel application of immobilized enzymes is presented. Cofactor dependent immobilized enzymes can be used as affinity chromatography supports to bind their substrates without alteration in absence of the cofactor. The procedure has been demonstrated using β-galactose dehydrogenase to retain D-galactose selectively in the absence of NAD. The uses of the technique are discussed from the down-stream processing and mechanistic points of view.
