Notes: The direct observation of crystal structure at ultra-low temperatures helps better understanding of phase transformations and phase stability. Two sets of 3He-4He dilution refrigerators with a top-loading facility were installed at BL-3C2 and 6C1 of the Photon Factory. The first is used exclusively for topography of quantum crystals of solid 3He and 4He for studies of lattice defects. Typical examples of topographs that indicate an annealing effect of solid helium, understood in terms of a quantum tunneling, are presented. The disappearance of downward diffraction Laue spots (2θB = 90°) is discussed on the basis of the temperature and other factors in solid helium. The second refrigerator is used for X-ray diffraction studies of crystalline solids. A splitting of the Laue spots from Cs2NaHoCl6 below 150 mK, giving evidence of a cooperative Jahn–Teller transition of the first kind at the lowest transition temperature ever observed, is briefly explained as an example of diffractometry.

Notes: Abstract. Objective: This study aimed to evaluate the abil-ity of S1319 (4-hydroxy-7-[1-(1-hydroxy-2-methylamino) ethyl]-1,3-benzothiazol-2(3H)-one acetate), a novel β 2-adrenoceptor selective agonist derived from marine sponge, to inhibit IgE-mediated activation of human cultured mast cells (HCMC) in vitro.¶Materials and Methods: We examined the effect of S1319 (racemate) on tryptase release and tumor necrosis factor- α (TNF-α) production in HCMC generated from human cord blood cells, after cross-linking of high affinity immunoglobulin E receptors (FcεRI), compared with those of the non-selective β-adrenoceptor agonist, isoproterenol (R-isomer), the selective β 2-adrenoceptor agonist, salbutamol (racemate), and the selective and long-acting β 2-adrenoceptor agonist, formoterol (racemate). We also evaluated the effect of S1319 on the intracellular cAMP level, inositol phosphate production and protein tyrosine phosphorylation in HCMC.¶Results: S1319 and β-adrenoceptor agonists inhibited the IgE-mediated release of tryptase. Approximate IC50 values of S1319, formoterol, isoproterenol and albuterol for the inhibition of tryptase release were 0.51 ± 0.12, 0.15 ± 0.1, 0.80 ± 0.09, and 28 ± 32.4 nM, respectively. S1319 and β-adrenoceptor agonists also inhibited TNF-α production by HCMC in a concentration-dependent manner. Approximate IC50 values of S1319, formoterol and isoproterenol for the inhibition of TNF-α production were 0.19 ± 0.03, 0.28 ± 0.02 and 0.32 ± 0.03 nM, respectively. S1319 caused a concentration-dependent increase in total cell cyclic AMP levels in HCMC. On the other hand, S1319 inhibited the accumulation of inositol 1,4,5-triphosphate and IgE-mediated protein tyrosine phosphorylation of 42-kDa protein, p42 mitogen activated protein (MAP) kinase (ERK-2).¶Conclusion: These results indicate that S1319 and β-adrenoceptor agonists are potent inhibitors of the IgE-mediated release of mediators from HCMC.

Notes: Abstract —We investigated the effects of various viscoelastic structures on postseismic surface displacement and principal strain fields associated with the great 1946 Nankaido earthquake, which occurred on the plate boundary between the subducting Philippine Sea plate and the continental Eurasian plate. For this purpose, we constructed two kinds of three-dimensional structural models using the finite element method one is the Layered Model, in which a semi-infinite Maxwell viscoelastic material is underlying an elastic layer, and the other is the more realistic Plate Model, in which the three-dimensional configuration of the subducted Philippine Sea plate is taken into account. We also considered two cases with different thicknesses of the elastic layer (50 and 33km) for the respective models. The difference between the two models in postseismic surface deformations is significant for the case with the thinner elastic layer. In this case the horizontal surface displacement and principal strain for the Layered Model is two to three times larger than those for the Plate Model. Downward surface deformation tends to be dominant for the Layered Model, while the change in the pattern for the Plate Model is less marked. The spatial extent of uplift and subsidence for the Plate Model is broader than that for the Layered Model. Postseismic vertical displacements in Shikoku were found to be strongly dependent on the viscoelastic structures. From these results, we suggest that the estimates of the viscosity of the uppermost mantle, interplate coupling, and the area and the amount of after-slip following the 1946 Nankaido earthquake, which have been estimated based on simple layered viscoelastic models, should be re-evaluated using realistic three-dimensionally heterogeneous viscoelastic structures.

Notes: Summary Heated albumin microspheres with an average diameter of 45±8 μm and containing mitomycin C, released, in vitro, about 20% of this antibiotic over a 3-day period. VX-2 tumors were implanted into the hind leg of rabbits and the drug-containing microspheres were injected into the femoral artery of these animals. High levels of the drug were maintained for several hours in the tumor and growth of the tumor was inhibited considerably, compared to findings in control rabbits given the conventional mitomycin C. Half the number of the rabbits treated with our new method are alive with no evidence of tumor.

Notes: Summary The cytotoxic activity of human peripheral blood lymphocytes against the natural killer-sensitive target K562 was suppressed both by prostaglandin E2 and dexamethasone. On the other hand, cultured lymphoid cells propagated in the presence of interleukin-2 showed strong cytotoxic reactivity against K562 targets, and were resistant to prostaglandin E2- or dexamethasone-mediated suppression.

Notes: Summary Vascular endothelial growth factor (VEGF) is a potent angiogenic factor. VEGF levels in ocular tissue of 6-, 12-, 18- and 28-week-old Goto-Kakizaki (GK) rats, a well-known model of non-insulin-dependent diabetes, were evaluated by highly sensitive ELISA. VEGF concentrations in the GK rat as well as in non-diabetic Wistar rat significantly decreased from the age of 6 weeks to 18 weeks. However, although VEGF concentrations in the Wistar rat continued to fall significantly from 18 to 28 weeks of age, the levels were maintained between 18 and 28 weeks of age in GK rats. Levels were significantly different between the GK and Wistar rats at 28 weeks of age. Results of immunohistochemical studies of the eyes of Wistar and GK rats at 28 weeks of age suggest diffuse distribution of this cytokine in cells of neural origin. Weak to moderate VEGF immunoreactivity was exhibited mainly in the ganglion cell layer, inner plexiform layer and inner/outer nuclear layers in rats with and without diabetes. However, in the retinal optic nerve fiber layer, retinal pigment epithelium and choroid, strong VEGF immunoreactivity was noted only in the GK rat. In conclusion, increased VEGF production in certain ocular tissue, similar to that in humans, is observed quite early, at least before the appearance of observable retinal changes in the diabetic GK rat. This also suggests that the GK rat can be used as a model of initial or latent phase diabetic retinopathy. [Diabetologia (1997) 40: 726–730]
