Summary
The isolation procedure of fibrin from human plasma and the analysis of the reduced protein in polyacrylamide gel electrophoresis in sodium dodecyl sulfate is described. Results concerning the subunit structure of highly purified fibrinogen, non cross-linked and completely cross-linked fibrin are presented.
It is demonstrated that the subunit structure depends on factor XIII-activity in plasma. Very low activities are sufficient to convert the γ-chains to the γ-dimer, while polymerization of the α-chains occurs only with factor XIII-activities within the normal range. Fibrin polymerization is a time-dependant reaction and is not finished before two hours of incubation at 37°C after adding thrombin and Ca++.
Fibrin of patients suffering from acute léukaemia and decompensated cirrhosis of the liver exhibited marked defects of the subunit structure without evidence of enhanced fibrinolytic activity in the blood.
Zusammenfassung
Die Isolierung von Fibrin aus Plasma und die Analyse der Fibrinstruktur in der PAA-Elektrophorese werden beschrieben. Die Ergebnisse der Untersuchung eines hochgereinigten Fibrinogenpräparates von nicht vernetztem und vollständig vernetztem Fibrin werden dargestellt. Es wird gezeigt, daß die Fibrinstruktur eng mit der Faktor XIII-Konzentration im Plasma korreliert ist. Während niedrige FSF-Aktivitäten zur Polymerisation der γ-Ketten ausreichen, kommt es erst bei hohen FSF-Aktivitäten zu einer vollständigen Polymerisierung der α-Ketten. Die Fibrinstabilisierung unterliegt darüber hinaus einer zeitlichen Kinetik und ist erst nach 2stündiger Inkubation bei 37°C vollständig abgeschlossen. Bei der Untersuchung von Gerinnseln aus Plasma von Patienten mit akuter Leukämie und dekompensierter Lebercirrhose zeigte sich eine defekte Fibrinstruktur ohne Hinweis auf eine Hyperfibrinolyse im Blut.
