Effect of several chemotherapeutic agents on protein synthesis in leukemic plasma cells

Summary

During the clinical course of a secondary plasma cell leukemia the cellular protein synthesis and immunoglobulin synthesis were measured in a cell-free system prepared from peripheral plasma cells. After treatment with cyclophosphamide and a combination of cyclophosphamide with melphalan and vincristine no change in the activity of total protein synthesis was found. In contrast, a decrease of total protein synthesis could be observed after treatment with BCNU combined with cyclophosphamide and Ara-C. The relative amount of 10–15% Ig-synthesis remained unchanged under the different chemotherapeutic regimes. These findings suggest a primary effect of BCNU on cellular protein synthesis in myeloma cells similar to that reported in HeLa cell cultures.
