DNA-dependent RNA polymerase activities and RNA synthesis as demonstrated by3H-uridin uptake are investigated after treatment of guinea pig epidermis with vitamin A-acid (VAA).

The local application of VAA results in a stimulation of the RNA polymerase activity over 100% after two applications. These effects are actinomycin D sensitive. After 6 and 8 days the effect of VAA on the RNA polymerase activity are less pronounced.

Also, after two applications with VAA (1%) the specific radioactivity of the isolated epidermal RNA was increased to about 60%.

Die Wirkung von Vitamin A-Säure (VAS) auf die Aktivität der DNS-abhängigen RNS-Polymerase in der Epidermis von Meerschweinchen und auf die epidermale RNS-Synthese mit Hilfe des Einbaus von3H-markiertem Uridin in die RNS wurde untersucht.

Die örtliche Applikation von VAS führt zu einer Stimulierung der RNS-Polymerase-Aktivität um 100–120% bereits 44 Std, d. h. nach zweimaliger Behandlung. Die Effekte von VAS sind Actinomycin-D empfindlich. Nach 6–8 tägiger Behandlung wird der VAS-Effekt auf die RNS-Polymerase-Aktivität kleiner.

Nach 2tägiger Behandlung mit einer 1%igen VAS-Lösung wurde eine Erhöhung der spezifischen Radioaktivität der isolierten epidermalen RNS gefunden.
