Different forms of the endoplasmic reticulum in neoplastic keratinocytes

Summary

Electron microscopic examination of squamous cell carcinoma of human epidermis showed different formations of the endoplasmic reticulum (ER) in neoplastic keratinocytes, which are unusual for normal epidermal cells. They can be characterized as follows:

  1. 1.

    Some cells show no ER and only few ribosomes.

  2. 2.

    Other cells demonstrate increased proliferation of the rough surfaced ER and the free polyribosomes.

  3. 3.

    One part of the cells shows a strong increase of free ribosomes, while scanty granulated tubes of rough surfaced ER are fragmented and diffusely distributed.

  4. 4.

    A hydropic dilatation of the ER is a predominant feature of some cells.

  5. 5.

    Smooth surfaced endoplasmic reticulum appears frequently, mostly arranged to systems of lamellae like “whorls” or “finger prints”. Alterationes of membranes can lead to cytolysis and to the formation of cytolysosomes.

  6. 6.

    Lipid droplets are recognized, more often in connection to the ER.

The morphological and also the functional abnormalities are not specific. The example of the neoplastic keratinocytes shows that malignant growth generally does not coincide with decreased cytoplasmic differentiation.

Zusammenfassung

Beim Plattenepithel-Carcinom menschlicher Epidermis zeigen entdifferenzierte Keratinocyten verschiedene Formationen des endoplasmatischen Reticulums (ER), die für normale Epidermiszellen ungewöhnlich sind. Sie lassen sich folgendermaßen charakterisieren:

  1. 1.

    Einige Zellen zeigen keinerlei ER und nur wenige Ribosomen.

  2. 2.

    Andere lassen eine starke Proliferation des rauhen ER und der freien Polyribosomen erkennen.

  3. 3.

    Ein Teil der Zellen zeigt eine starke Vermehrung freier Ribosomen, während spärlich granulierte ER-Schläuche eine Fragmentation und diffuse Verteilung aufweisen.

  4. 4.

    Eine hydropische Dilatation des ER beherrscht das Zellbild.

  5. 5.

    Glattes ER tritt gehäuft auf, oft als „whorls“ oder „finger prints“ zu Lamellensystemen geordnet. Membranschäden können zur Cytolyse und zur Bildung von Cytolysosomen führen.

  6. 6.

    In Beziehung zum ER lassen sich vermehrt Lipidtropfen erkennen.

Die morphologischen und damit auch funktionellen Abweichungen von der Norm sind unspezifischer Natur. Sie zeigen am Beispiel der malignen Keratinocyten, daß malignes Wachstum nicht generell mit einer Abnahme der Cytoplasmadifferenzierung einhergeht.
