Chromosomenuntersuchungen an menschlichen Lymphocytenkulturen nach Zugabe von Cyproheptadin

Abstract

Human lymphocyte cultures were exposed to different concentrations of cyproheptadine (0.55×10⁻⁴; 1.1×10⁻⁴; 2.2×10⁻⁴; 4.4×10⁻⁴M) for 24 and 5 hrs before the end of incubation time and examined for induced chromosomal damage. With the concentration 4.4×10⁻⁴ M, as well as in the 24-hour test with a concentration of 2.2×10⁻⁴ M, the toxic damage was so severe that only few, greatly deformed cell nuclei were found. 300 metaphases each for the 5-hour exposure with 2.2×10⁻⁴ M and for the lower concentrations could be evaluated. No increase of chromosomal damage was observed in comparison to the controls. These results are discussed in the context of other findings.

Zusammenfassung

Menschliche Lymphocytenkulturen wurden verschiedenen Konzentrationen von Cyproheptadin (0,5×10⁻⁴; 1,1×10⁻⁴; 2,2×10⁻⁴; 4,4×10⁻⁴ M) für jeweils 24 und 5 Std vor Ende der Inkubationszeit ausgesetzt und auf induzierte Chromosomenschädigungen untersucht. Bei der Konzentration 4,4×10⁻⁴ M sowie bei dem 24 Std-Versuch mit der Konzentration 2,2×10⁻⁴ M war die toxische Schädigung so stark, daß nur vereinzelte, zum großen Teil deformierte Zellkerne gefunden wurden. Mit je 300 ausgewerteten Metaphasen konnten nur das 5 Std-Experiment mit 2,2×10⁻⁴ M und die niedrigeren Konzentrationen beurteilt werden. Es fand sich keine Erhöhung der Chromosomenschädigungen gegenüber den Kontrollen. Die Ergebnisse werden im Vergleich mit anderen Befunden diskutiert.
