Summary
Intraperitoneal injection of PGE1 (0.1–1.0 mg/kg) in fasting rats produced a long-lasting and dose-dependent increase in blood sugar but showed no influence upon the level of serum free fatty acids.
The catecholamine content of the adrenal glands was significantly decreased 60 and 120 min after the intraperitoneal injection of PGE1 (0.5–1.0 mg/kg). PGE1 in vitro did not liberate any histamine from isolated peritoneal mast cells of the rat.
The glycogen content of rat liver slides was significantly decreased by incubation of the tissuein vitro with PGE1 (75–100 μg/ml) for 30 min at 37° C.
The results favour the assumption that the increase in blood sugar induced by PGE1 is the result of first a direct glycogenolytic effect of PGE1 within the liver and second an indirect action by liberation of catecholamines from the adrenal glands.
Zusammenfassung
Intraperitoneale Injektion von PGE1 (0,1–1,0 mg/kg) führte bei Ratten im Hungerzustand zu einer lange anhaltenden und dosisabhängigen Blutzuckersteigerung. Die freien Fettsäuren im Serum wurden hingegen nicht beeinflußt.
Nach intraperitonealer Injektion von 1,0 mg/kg PGE1 trat in den Nebennieren der Tiere eine kontinuierliche Verminderung des Gesamtbrenzkatechinamingehalts ein, die sich nach 60 und 120 min als signifikant erwies.In vitro setzte PGE1 (10–100 μg/ml) aus isolierten Mastzellen der Ratte kein Histamin frei.
Nach 30 min Inkubation mit PGE1 (75–100 μg/ml) nahm der Glykogengehalt von Rattenleberschnittenin vitro gegenüber den Kontrollen signifikant verstärkt ab.
Auf Grund der Ergebnisse wird angenommen, daß die nach PGE1-Applikation auftretende Hyperglykämie ihre Ursache erstens in einem direkten glykogenolytischen Effekt der Substanz in der Leber und zweitens in einer indirekten, über Brenzkatechinaminfreisetzung aus dem Nebennierenmark verlaufenden, Wirkung hat.
