Zusammenfassung
Die Anpassung eines Columbia-SK-Virusstammes an L-Zellen wurde vergleichend mit fluoreszenzserologischen und virologischen Methoden untersucht. Mit der Immunfluoreszenztechnik können in den einzelnen Passagen Informationen über den Anteil der infizierten Zellen, die Fluoreszenzintensität und die Verteilung des Antigens in der Zelle gewonnen werden.
Auf Grund des Infektiositätstiterverlaufes, des Anteils fluoreszierender Zellen und der Änderung der Plaquegrößen während der Adaptation ist zu schließen, daß die Anpassung das Ergebnis einer Selektion ist, der vermutlich mehrere Mutationsschritte vorausgingen. Der an L-Zellen angepaßte Virusstamm wird mit Columbia-SKL, bezeichnet.
Summary
The adaptation of a Columbia-SK virus strain to L cells was investigated both by means of fluorescent antibodies and virological methods. Using the immunofluorescence technique it is possible to follow the percentage of infected cells as well as the intensity of fluorescence and the distribution of the antigen within the cells during the successive passages of the virus.
By reason of the infectivity titers, the proportion of fluorescent cells, and the changes in plaque sizes during the consecutive virus passages it is proposed that a selection of presumedly several mutational steps is the cause of the adaptation observed. The virus adapted to L cells is designated as Columbia- SK4 virus.
