Zusammenfassung
1. Die Cholesterinsyntheserate in der Leber aminonucleosid-nephrotischer Ratten wurde durch den Einbau von14C-Acetat bzw.14C-Mevalonat am intakten Tier bestimmt. Gleichzeitig wurde das Erscheinen von14C-markiertem Cholesterin im Plasma gemessen.
2. Zwischen dem 10. und 12. Tag nach Beginn der Aminonucleosidinjektion läßt sich eine starke Steigerung der hepatischen Cholesterinsynthese aus14C-Acetat nachweisen: Gegenüber den Kontrolltieren ist die spezifische Aktivität des Lebercholesterins um das Zwei- bis Vierfache erhöht.
3. Am 13.–15. Tag, bei ansteigendem Plasmacholesterin, tritt eine Hemmung der Cholesterinsynthese ein, kenntlich am Absinken der spezifischen Aktivität und des absoluten Einbaus unter die entsprechenden Kontrollwerte.
4. Bei Verwendung von14C-Mevalonat ist der Anstieg nur wenig auspeprägt, eine Einbauhemmung in den späteren Nephrosestadien ist ebenfalls nicht nachweisbar.
5.|Die Versuchsergebnisse werden mit den Daten anderer Autoren verglichen und im Zusammenhang mit den derzeitigen Kenntnissen über die Regulation der Cholesterinbiosynthese diskutiert.
Summary
1. The rate of cholesterol formation in the liver of rats with experimental nephrosis was studied by incorporation of14C-acetate and14C-mevalonate in the whole animal. Plasma levels of labelled cholesterol were also measured.
2. 10 to 12 days after administration of aminonucleoside hepatic cholesterol synthesis from14C-acetate is clearly enhanced: the specific activity of liver cholesterol is 2–4 times increased as compared with control animals.
3. On the 13. to 15. day with increasing plasma cholesterol an inhibition of cholesterol synthesis is observed by the specific and total activities decreasing below the control values.
4. On administration of14C-mevalonate the specific activity of cholesterol increases only 20 to 50%, an inhibition of cholesterol formation in the later stages of nephrosis cannot be shown.
5. These results are compared with data of other authors and discussed with regard to the present knowledge of regulation of cholesterol biosynthesis.
