The influence of adrenergic receptor-blocking agents, amphetamine, and 6-aminonicotinamide on thermoregulation

Summary

The influence ofα- andΒ-receptor-blocking agents, as well as that of amphetamine and of 6-aminonicotinamide (6-AN) on the body temperature of the rat was examined at three different ambient temperatures (AT). The ambient temperature is crucial both for the direction and the potency of the caloric effect. The ambient temperature at which a drug does not affect the body temperature is referred to as the specific indifferent temperature. At + 4‡C and + 22–24‡C., the α-receptor blockers phentolamine, and Dihydroergotamine (DHE), as well as 6-AN lower the body temperature, while theΒ-receptor blockers D(−)INPEAandL(+)INPEA do not interfere with it. The specific indifferent temperature of amphetamine lies below 22‡C. Higher ambient temperatures result in hyperthermia, lower temperatures lead to hypothermia. The results obtained in combined treatment demonstrate that the thermic effect produced by amphetamine at + 4‡C AT probably does not develop via adrenergic mechanisms. In addition, our studies do not rule out the possibility that 6-AN may influence body temperature in a similar way as phentolamine and DHE.
