Summary
Solutions of high molecular calf-thymus-DNA were mixed with dimethyl formamide solutions of 12 cancerogenic inactive and active aromatic hydrocarbons and amines in order to study the reciprocal effect with DNA. The solutions were then dialyzed, extracted with organic solvents, and centrifuged. From the UV spectra of the solutions the following were determined: (a) the amount of “bound” compounds from the bands of aromatics above 300 mμ; (b) the extinction at 260 mμ as a measure of the DNA present in the solution. By following the extinction with increases in temperature the hyperchrome effect and the “melting point” of DNA were determined. Relationships between the data obtained and the cancerogenic activity of the compounds were not found. In addition, pyridino-[2,3∶3,4]-phenoxazon-(2) and the follicle hormone estradiol-(3,17 β) were without effect. The reciprocal effect of DNA with the mutagens, proflavin and acridine orange, was confirmed. A distinct increase of the “melting point” of DNA by means of N-phthalyl-glutaminic acid-imide was striking.
