Zusammenfassung
16-Methylen-14C-prednisolon, ein synthetisches Steroid mit überwiegender Glucocorticoidwirkung, wurde sieben Normalpersonen intravenös oder peroral verabreicht und die biologischen Eigenschaften dieses Hormons bestimmt. Nach intravenöser Injektion des radioaktiven Steroids war die Schwundrate des Hormons bei vier Normalpersonen bis 4 Std p.i. größer als in der Zeit von 4–8 Std p.i. Aus der Plasmakonzentration des Hormons errechnete sich eine biologische Halbwertszeit von 161±32 min. Das fiktive Verteilungsvolumen, berechnet aus der Konzentration des unveresterten Hormons im Gesamtplasma zur Zeitt 0 war in der Zeit von 60–240 min p.i. 25,8–52,81 und zwischen 240–480 min p.i. 61,9 bis 189,61. Nach peroraler Verabreichung des Hormons wurde bei drei Versuchspersonen die maximale Plasmakonzentration von 1,1–1,7% der applizierten Dosis pro 1000 ml Plasma nach 120–200 min erreicht. Im Urin wurden in den ersten 24 Std p.i. nach intravenöser Verabreichung von 16-Methylen-14C-prednisolon im Mittel 90,7% der injizierten Aktivität ausgeschieden; nach peroraler Gabe waren es im Mittel 86,0%. Die Ausscheidung von unveresterten radioaktiven Steroiden war mit im Mittel 26% der injizierten Dosis relativ groß. Als Glucuronsäureester wurden im 24 Std-Urin nur 24% nachgewiesen. 7,6% des injizierten Steroids wurden unverändert als freies 16-Methylen-14C-prednisolon im Urin gefunden. Neben 16-Methylen-14C-prednisolon und 16-Methylen-14C-prednison wurden in den Urinextrakten noch sieben radioaktive Metaboliten nachgewiesen. Diskutiert wurden die Zusammenhänge zwischen chemischer Struktur und biologischen Eigenschaften von Steroidhormonen.
Summary
16-methylen-14C-prednisolon a synthetic steroid with the predominant effects of a glucocorticoid was given 7 normal subjects intravenous or orally and the biological properties of the hormone were estimated. After intravenous injection of the radioactive steroid the disappearance rate was 1–4 hours after the injection greater than between 4 and 8 hours. The biological half-time of 16-methylen-14C-prednisolone was calculated as 161±32 min. The volume of distribution was 60–240 min after the injection 25.8–52.81 and between 240–480 min 61.9–189.61. After oral application of the steroid the maximum of the plasma concentration of 1.1–1.7% of the dose injected was reached within 120–200 min. In urine we found after intravenous injection of 16-methylen-14C-prednisolone 90,7% of the activity and after oral application 86%. 26% of the injected steroid were excreted as unesterified metabilities of 16-methylen-14C- prednisolone and 24% as glucuronides. 7.6% of the injected steroid we found unchangen in urine. Besides 16-methylen-14C-prednisolone and 16-methylen-14C-prednisone 7 radioactive metabilities were demonstrable in urine.
