Summary
Five members of a German family showed a congenital haemolytic anaemia which is characterized by the occurrence of large inclusion bodies in the erythrocytes of the patients.
These inclusion bodies were demonstrable by means of Nile blue sulfate stain-even before splenectomy in a degree up to 40%.
The origin of the anaemia should be detected as an abnormal heat-labile haemoglobin.
This unstable haemoglobin precipitates even as CO-haemoglobin; spectroscopic and electrophoretic analyses show that it is different from Hb Köln.
Based on the clinical symptoms in the carriers of the inherited defect and on the different properties of the abnormal haemoglobin it was assumed that this was a new unstable haemoglobin variant; it was named “Hb Frankfurt”.
Zusammenfassung
Bei fünf Angehörigen einer deutschen Sippe wurde eine kongenitale hämolytische Anämie beobachtet, die dadurch charakterisiert ist, daß in den Erythrocyten der Betroffenen in einem Ausmaß bis zu 40% große, grobschollige Innenkörper auftreten, die bereits vor Splenektomie färberisch mit Nilblausulfat nachweisbar sind.
Als Ursache dieser Anämie konnte ein anomales hitzelabiles Hämoglobin festgestellt werden. Dieses instabile Hämoglobin, das selbst als CO-Hb leicht präcipitiert, verhält sich elektrophoretisch und bei der Spektralanalyse anders als Hb Köln. Auf Grund der klinischen Symptomatik bei den Erbmalsträgern und auf Grund der besonderen Eigenschaften des anomalen Hb wird angenommen, daß es sich um eine neue instabile Hb-Mutante handelt; sie wurde „Hb Frankfurt“-benannt.
