Summary
Our experiments carried out on the isolated perfused rat pancreas show that: 1. In the absence of glucose, the isolated perfused rat pancreas secretes only a very small amount of insulin; which is termed the basal secretion. — 2. When increasing concentrations of glucose are used, the insulin output rate increases as a function of the concentration of glucose (following a sigmoidshaped curve). — 3. Tolbutamide alone without glucose slightly increases the basal insulin secretion rate. — 4. Tolbutamide potentiates the insulin secretory effects of increasing concentrations of glucose: in the presence of tolbutamide at 100 mg/1, the additional amount of secreted insulin rises as the glucose concentration rises, up to 2 g/1; at 3 g/1 it decreases. — 5. At high concentrations (100 mg/1), the potentiating effect of tolbutamide on insulin secretion due to glucose (1.5 g/1) persists long after the termination of the sulphonamide infusion. At a lower concentration (5 mg/1) it ceases when the sulphonamide infusion is stopped. — 6. Glibenclamide (at low dosage: 1 μg/1) not only potentiates the action of glucose, but also prolongs its stimulating action on the beta cell, even long after the sulphonylurea has been removed from the perfusion fluid. — 7. In our experimental conditions, repeated tolbutamide administration in the presence of glucose, does not modify the insulin secretory response of the pancreas to the sulphonamide.
Résumé
Nos expériences effectuées sur le pancréas isolé et perfusé du rat montrent que: 1. En l'absence de glucose, le pancréas sécréte seulement une trés petite quantité d'insuline: c'est la sécrétion basale. — 2. Quand on utilise des concentrations croissantes de glucose, le débit d'insuline augmente en fonction de la concentration de glucose (suivant une courbe sigmoïde). — 3. Le tolbutamide seul, en l'absence de glucose, augmente légérement la sécrétion basale. — 4. Le tolbutamide potentialise les effets insulino-sécréteurs de concentrations croissantes de glucose. En présence de tolbutamide, la quantité supplémentaire d'insuline sécrétée s'éléve en même temps que la concentration de glucose, jusqu'à 2 g/1; à 3 g/1 elle diminue. — 5. A forte concentration (100 mg/1), l'effet potentialisateur du tolbutamide sur l'insulino-sécrétion due au glucose (1.5 g/1) se prolonge bien au delà de l'arrêt de l'infusion du sulfamide. A faible concentration (5 mg/1) il cesse dés que l'infusion est supprimée. — 6. Le glibenclamide (à faible concentration: 1 μg/1), non seulement potentialise l'action du glucose, mais aussi prolonge son action stimulante sur la cellule bêta, longtemps aprés la suppression du sulfamide hypoglycémiant. — 7. Dans nos conditions expérimentales, l'administration répétée de tolbutamide, en présence de glucose, ne modifie pas la réponse insulino-sécrétoire du pancréas à ce sulfamide.
Zusammenfassung
Unsere Untersuchungen am isolierten, perfundierten Rattenpankreas zeigen: 1. In Abwesenheit von Glucose sezerniert das Pankreas nur eine sehr kleine Insulinmenge: Sie entspricht der Basalsekretion. — 2. Bei Verwendung steigender Glucosekonzentration steigt die Insulinausschüttung in Abhängigkeit vonder Glucosekonzentration an (sie beschreibt dabei eine S-förmige Kurve). — 3. Tolbutamid allein ohne Glucosezusatz steigert die Basalsekretion geringfügig. — 4. Das Tolbutamid potenziert die Wirkung steigender Glucosekonzentrationen auf die Insulinsekretion. In Gegenwart von Tolbutamid steigt die zusätzlich sezernierte Insulinmenge parallel zur Glucosekonzentration bis 200 mg% an; ab 300 mg% verringert sie sich. — 5. Bei hohen Tolbutamidkonzentrationen (100 mg/1) hält sein potenzierender Effekt auf die durch Glucose (150 mg%) stimulierte Insulinausschüttung noch lange nach Beendigung der Sulfonamidinfusion an. Bei niedrigen Konzentrationen (5 mg/1) hört er bei Abbruch der Infusion sofort auf. — 6. Schon niedrige Konzentrationen von Glibenclamid (1 μg/1) verstärken nicht nur den Effekt der Glucose, sondern verlängern auch ihre stimulierende Wirkung auf die B-Zelle noch lange über die Beendigung der Zufuhr des blutzuckersenkenden Sulfonamids hinaus. — 7. Unter unseren Versuchsbedingungen verändert die wiederholte Zufuhr von Tolbutamid in Gegenwart von Glucose die Reaktion der Insulinausschüttung des Pankreas auf dieses Sulfonamid nicht.
