Die heterotrophe C14O2-Assimilation von Ochromonas malhamensis führt hauptsächlich zur Markierung von Säuren und Nebenprodukten des Citronensäurecyclus und von Alanin und Milchsäure. Von den möglichen Enzymen zur Carboxylierung von C3-Körpern sind wahrscheinlich das zu Äpfelsäure führende “malic enzyme” und die Oxalacetat bildenden Fermente PEP-Carboxykinase oder Pyruvat-Carboxylase nebeneinander vorhanden.

Bei thiaminverarmten Ochromonas-Zellen ist das osmotische Gleichgewicht gestört.

Bei Thiaminmangel ist die heterotrophe CO2-Assimilation stark vermindert. Dazu kommt es hauptsächlich durch eine Hemmung der von malic enzyme katalysierten Carboxylierung. Der Anteil von Alanin und Milchsäure an der Radioaktivität fällt ab, und der Anteil von Glutaminsäure und Citronensäure nimmt stark zu. Diese Effekte werden aus der Hemmung der oxydativen Decarboxylierung von Pyruvat und α-Ketoglutarsäure erklärt.

Die Dunkelfixierung von CO2 ist bei Biotinmangel durch Inaktivierung der PEP-Carboxykinase und/oder der Pyruvat-Carboxylase stark vermindert. Die geringe Oxalacetatbildung äußert sich vor allem in einem fast vollständigen Verschwinden des Anteils der radioaktiven Asparaginsäure. Die restliche Dunkelfixierung erfolgt ausschließlich durch das malic enzyme.
