Functional importance of sulfhydryl groups in membranes of lysosomal granules of bovine polymorphonuclear leucocytes

Summary

A number of mercury sulfhydryl-reagents act on isolated lysosomal granules of bovine polymorphonuclear leucocytes in concentrations of 10−3 to 10−7 M/l to cause a decrease of optical density and the liberation of acid phosphatase, cathepsin and cationic polypeptides.

Electron microscopy of isolated granules after treatment with mersalyl showed a decrease in electron density of the matrix and vacuolisation. Concentrations up to 10−3 M/l destroyed the particles and ruptured their membranes. The number of sulfhydryl groups was estimated at 1 Mol SH per 22000 g of granule protein. Blocking these groups with p-chlormercuribenzoate causes an increase in membrane permeability for enzymes of the lysosomal particles.
