Zusammenfassung

Es wird über die Befunde quantitativerFeulgen-photometrischer DNA-Bestimmungen an 30 Kranken mit Paramyeloblastenleukämie und 15 Kranken mit chronisch-myeloischer Leukämie berichtet. Von den 15 Kranken mit chronisch-myeloischer Leukämie konnten 8 im Myeloblastenschub und 6 im terminalen Stadium eines Myeloblastenschubes untersucht werden.

Auf Grund des DNA-Gehaltes der Paramyeloblasten bzw. der Granulozytenvorstufen bei chronisch-myeloischer Leukämie läßt sich der Anteil der in DNA-Synthese befindlichen Zellen bestimmen.

Bei 29 der 30 Paramyeloblastenleukämien fand sich ein signifikant erniedrigter Anteil Paramyeloblasten in DNA-Synthese.

Ein Unterschied im Prozentsatz der in DNA-Synthese stehenden Paramyeloblasten zwischen der Untersuchung a) am peripheren Blut und Sternalmark, b) zwischen vorbehandelten und unbehandelten Kranken, c) bei leukopenischen und leukozytotischen Verlaufsformen, d) im finalen Stadium, Untersuchung innerhalb 14 Tagen vor dem Tod, ließ sich nicht wahrscheinlich machen. Mit einem P von 0,08% ist nur eine Abnahme der DNA-synthetisierenden Zellen mit zunehmender Krankheitsdauer in den ersten Wochen der Erkrankung wahrscheinlich.

Bei den teilungsfähigen Granulozytenvorstufen unbehandelter und behandelter Kranker mit chronisch-myeloischer Leukämie fand sich ein Anteil in DNA-Synthese stehender Zellen wie in der normalen Granulopoese.

Der Anteil der DNA-synthetisierenden Zellen lag in den Myeloblasten im Myeloblastenschub bei chronisch-myeloischer Leukämie im Mittel signifikant niedriger als in der normalen Granulopoese und am deutlichsten erniedrigt im finalen Myeloblastenschub. Er lag jedoch noch signifikant über dem Mittelwert der 30 untersuchten Kranken mit Paramyeloblastenleukämie.

Die Befunde einer verlängerten Generationszeit leukämischer Zellen werden im Hinblick auf die in den meisten Fällen ausgeprägte Zellvermehrung diskutiert.

The findings of quantitative Feulgen photometric DNA determinations on 30 patients with paramyeloblastic leukaemia and 15 patients with chronic myeloid leukaemia are reported. Of the 15 patients with chronic myeloid leukaemia 8 could be examined during a myeloblastic exacerbation and 6 during the terminal stage of such an exacerbation.

On the basis of the DNA content of the paramyeloblasts or the granulocyte precursors respectively, in cases of chronic myeloid leukaemia, the portion of cells synthetising DNA can be determined.

In 29 out of 30 cases of paramyeloblastic leukaemia a significantly diminished portion of paramyeloblasts were found to be synthetising DNA.

A difference in the percentage of paramyeloblasts synthetising DNA between the examinations of a) peripheral blood and sternal marrow, b) treated and untreated patients c) leukopenic and leucocytotic forms, d) and during the final stage, with examination conducted within 14 days of death, could not be demonstrated. With a P of 0.08%, a decrease of DNA synthetising cells with increasing duration of the disease, is only probable during the first weeks of the disease.

In the granulocytic precursors capable of division of untreated and treated patients with chronic myeloid leukaemia, a portion of cells synthetising DNA was found to be the same as in normal granulopoesis.

The proportion of cells synthetising DNA in chronic myeloid leukaemia in myeloblasts during a myeloblastic exacerbation was in the mean significantly lower than in normal granulopoesis and most markedly reduced in the terminal myeloblastic exacerbation. It was, however, significantly higher than the mean value in 30 patients examined with paramyeloblastic leukaemia.

The various findings are discussed and it is attempted to explain the cell increase, which is marked in most cases.
