Zusammenfassung
Die Seren von 50 Blutspendern wurden zum Nachweis des C-reaktiven Proteins (CRP) mit einem handelsüblichen Anti-CRP-Serum mit Immunodiffusionstechniken im Agargel getestet. Dabei konnte in 72% aller Seren bei Verwendung des Anti-CRP-Serums in einer Verdünnung von 1∶16 eine Präzipitationslinie nachgewiesen werden. In den übrigen 28% der Seren wurde das CRP entweder durch Einengung der Seren oder Verwendung höherer Anti-CRP-Verdünnungen erfaßt. Bei Vergleich dieser Linie mit dem spezifischen Präzipitat von 25 CRP-positiven Seren ergab sich immunologische Identität der beiden Linien. Die immunoelektrophoretische Position des CRP-Präzipitats der normalen Seren entsprach der der CRP-positiven Seren. Mit Hilfe der einfachen radialen Immunodiffusion konnte gezeigt werden, daß die CRP-Konzentration der Normalseren in etwa um das 8-bis 10fache niedriger liegt als die CRP-Konzentration in CRP-positiven Patientenseren.
Die Untersuchungsergebnisse machen es wahrscheinlich, daß das CRP ein physiologisches Serumprotein darstellt, dessen Funktion in einer initialen, unspezifischen Reaktivität im Rahmen von Abwehrvorgängen infektiöser oder entzündlicher Prozesse besteht; es wird offenbar abgelöst mit dem Ingangkommen der spezifischen Antikörperbildung.
Summary
The sera of 50 blood donors were analysed for C-reactive protein (CRP) using immunodiffusion techniques in agargel with a commercially available anti-CRP-serum. In 72% of all sera a precipitation line could be shown using the anti-CRP-serum in a dilution of 1∶16. In the other 28% of normal sera CRP has been comprehended either by concentrating the sera or by higher dilutions of anti-CRP-serum. Comparing this precipitate with the specific line of 25 CRP-positive sera identical immunological reactions were found in all. The immunoelectrophoretic position of the CRP precipitate in normal sera corresponded to that of CRP-positive sera. By quantitative immunodiffusion it could be demonstrated that on average the CRP concentration in normal sera was eight to ten times lower than in CRP-positive sera. We suppose that CRP is a new class of normal serum proteins which probably acts as an unspecific factor at the very beginning of immune response of infectious diseases or inflammatory processes; it obviously disappears in the phase of specific antibody production.
