Abstract
The binding of calcium ions to purified elastin was shown to be dependent on pH and the Ca2+ concentration of the medium; such binding occurs only above pH 5.5 and it increases with increasing pH. These data may be explained by assuming electrostatic interaction between Ca2+ and ionised groups of the organic matrix, probably carboxyl-and amino-groups located in some polar areas along with hydrophobic structure of the elastin macromolecule. No binding of phosphate was observed at physiological pH.
Résumé
La fixation d'ions calcium sur de l'élastine purifiée dépend du pH et de la concentration du milieu en Ca2+. Une telle fixation ne s'observe qu'au dessus de pH 5,5 et augmente avec l'élévation du pH. Ces résultats peuvent s'expliquer en admettant une interaction électrostatique entre Ca2+ et des groupements ionisés de la matrice organique, probablement des groupes carboxyles et aminés, situés dans certaines régions polaires, ainsi qu'à la structure hydrophobique de la macromolécule d'élastine. Aucune fixation de phosphate n'est observée à pH physiologique.
Zusammenfassung
Es wurde gezeigt, daß die Bindung von Calciumionen an gereinigtes Elastin vom pH und von der Ca2+-Konzentration des Mediums abhängig ist; eine solche Bindung findet nur bei einem pH über 5,5 statt und nimmt bei steigendem pH zu. Diese Feststellung kann durch die Annahme erklärt werden, daß eine elektrostatische Wechselwirkung vorliegt zwischen Ca2+ und den ionisierten Gruppen der organischen Matrix, vermutlich Carboxyl- und Aminogruppen, welche an einigen polaren Stellen auftreten, verbunden mit der hydrophoben Struktur des Elastinmakromoleküls. Bei einem physiologischen pH konnte keine Phosphat-bindung beobachtet werden.
