Abstract

Farnesal and 3-hydroxy-2,3-dihydrofarnesal (3-hydroxy-3,7,11-trimcthyl-6,10-dodecadiene-1-al) were formed from farnesol when the alcohol was incubated with the protoplast of Botryococcus braunii B race strain. This fact suggests the existence of farnesal hydratase in the alga. Feeding experiments showed that both farnesal and 3-hydroxy-2, 3-dihydrofarnesal were efficiently incorporated into botryococcenes, triterpenoid hydrocarbons of the alga.

On the other hand, it is also plausible that degradation of dolichol is involved in determining its cellular concentration. The mechanism of dolichol catabolism remains unknown even though its half life has been estimated in animal tissue (t½ 40–70 h and 80–140 h in sea urchin embryo and rat liver, respectively [36,37]), and oxidative breakdown via conversion to aldehyde [38,39] or carboxylic acid [40,41] has been postulated. The polyisoprenoid alcohols (predominantly dolichols) accumulated in the Arabidopsis thaliana hairy roots occurred as esters of short-chain carboxylic acids, mainly propionic acid, and no free dolichols were detected at any time-point studied.

According to the above results, glucose was selected as carbon source and its optimal initial concentration was 2.5 g L−1. Here KNO3 was used as nitrogen source in 250 mL flasks according to the literature [31]. The initial concentrations of KNO3 were 0.4 g L−1, 0.8 g L−1, 1.0 g L−1, 1.2 g L−1 and 1.6 g L−1 respectively, and the inoculum was 80 g m−3.

The contents in the vials were completely dried and dissolved in 500 μl of H2O, and the incorporation of radioactivity was measured with a scintillation counter. Previous in vivo precursor feeding studies on the synthesis of botryococcene have suggested that FPP may not be a substrate for a putative BS enzyme [23,24]. To evaluate this possibility, an aliquot of a 2000g supernatant prepared from a B. braunii cell homogenate was incubated at 37 °C for 30 min in the presence of [3H]FPP, MgCl2, and NADPH.
