Retention of messenger activity of RNA phage RNA following RPC-5 chromatography

Abstract

RNA from bacteriophages MS2 and Qβ can be chromatographed on reverse-phase-chromatography-5 (RPC-5) columns and, subsequent to the chromatography, the RNAs retain their messenger activity and direct the initiation of their respective gene products in approximately the proper relative proportions. Further, MS2 RNA chromatographs as a discrete peak even in the presence of bulk Escherichia coli RNA on these columns. Pulse-labeled E. coli RNA is spread throughout the fractions of an RPC-5 column separation, suggesting that individual bacterial messenger RNA species could be fractionated in this manner. These results indicate that RPC-5 column chromatography is a potentially useful procedure as a method for fractionating and purifying mRNA.
