Abstract
OF the chemical substances that possibly mediate the inflammatory response1,2, bradykinin (H–L-Arg–L-Pro–L-Pro–Gly–L-Phe–L-Ser–L-Pro–L-Phe–L-Arg–OH) warrants special study because four of its pharmacological activities—the capacity to increase vascular permeability and to initiate vasodilation, pain and the emigration of leucocytes—are all encountered in acute inflammation3. We have sought a correlation of the structure of bradykinin with its ability to increase vascular permeability. The effect on activity was investigated when each of the nine amino-acids in the peptide chain was replaced in turn with L-alanine.
