Zusammenfassung
Der cytochemische Nachweis von Esterasen hat in der Hämatologie eine unbestreitbare Bedeutung erlangt. Die Darstellung esterolytischer Aktivitäten an Ausstrichen und Gewebsschnitten erlaubt eine wesentlich bessere Identifizierung bestimmter Zellformen, als sie mit den panoptischen Färbungen möglich ist. Eine weitgehend spezifische Darstellung der neutrophilen Granulocyten und deren Vorstufen ergibt die Naphthol-AS-D-Chloroacetat-Esterase; Monocyten lassen sich im Ausstrich sowie in Infiltraten und Exsudaten mit Hilfe einer Doppelinkubation zum Nachweis NaF-hemmbarer und -unhemmbarer Naphthol-AS-Acetat-Esterase eindeutig identifizieren.
Der sichere Nachweis von Monocyten und Monocytenvorstufen im Knochenmark festigt die Hypothese der myeloischen Herkunft dieser Zellen. Für die Diagnose der Monocytenleukämie ist der Nachweis der durch NaF hemmbarer Esterasen von entscheidender Bedeutung. Zur weiteren Differenzierung akuter Leukämien kann der Nachweis der Naphthol-AS-D-Chloracetat-Esterase (promyelocytäre Formen) herangezogen werden.
Summary
The cytochemical demonstration of esterases in hematologic studies is now well established. Demonstration of esterolytic activities on tissue sections and smears of blood and bone marrow allows an easier identification of various cell types. Monocytes can be identified in blood and marrow smears as well as in infiltrates and exudates with a double staining technique for the demonstration of NaF-sensitive and NaF-resistant esterase. With the aid of the naphthol-AS-D-chloroacetate-esterase reaction a fairly specific characterization of neutrophile granulocytes and their precursors is achievable.
The reliable identification of monocytes and monocytic precursors in the bone marrow confirms the hypothesis of the myeloid origin of these cells. For the diagnosis of monocytic leukemia the demonstration of a considerable amount of NaF-se sitive naphthol-AS-esterase in the leukemic cells is d cisive. Further differentiation of acute leukemias can be achieved with the aid of the naphthol-AS-D-chloroacetate-esterase reaction (promyelocytic types).
