Zusammenfassung
Stoffwechselintensive Streptomyceten zeigen bei extremer Stickstoffbelastung eine starke Pyruvatstauung, die durch eine verminderte Decarboxylierung von glykolytischer Brenztraubensäure ausgelöst wird. Ursache dafür ist eine erhöhte Verstoffwechslung von α-Ketoglutarsäure infolge einer konkurrierenden Inanspruchnahme der Co-Faktoren. Wir nehmen an, daß die Beanspruchung und Neusynthese der Co-Faktoren erst nachträglich regulativ koordiniert werden. Dieser Pyruvatstau führt aber keineswegs zum Stoffwechselstillstand, sondern wahrscheinlich durch Hemmung im unteren Glykolysebereich, zu einer anhaltenden Metabolitstauung und damit zu einer Aktivierung des Hexosemonophosphatweges. Die Nachschubreaktion wird damit beeinträchtigt, und das angehäufte Pyruvat wird wieder abgeführt. Auf diese Weise funktioniert der HMP als regulativer Ausweichstoffwechsel. Gleichzeitig fanden wir bei α-Ketoglutarsäurebelastung eine selektive Anhäufung von Serin (?), was wir als vermutliches Reaktionsprodukt einer Aminierung zwischen α-Ketoglutarsäure und Oxalessigsäure (im TCC) diskutieren.
