Sensitive quantitation of unlabelled SV40 cRNA by competition hybridization with3H-labelledin vitro cRNA

Summary

A sensitive method has been developed for the quantitation of small amounts of SV40 cRNA by means of competition hybridization of3H-SV40 cRNA (transcribed in vitro with RNA polymerase fromE. coli). The rate of competition is only slightly diminished by a moderate excess of filter-bound SV40 DNA. By using a tenfold excess of SV 40 DNA and a3H-SV40 cRNA of high specific activity it is possible to obtain a competition rate of 50% with only ng amounts of unlabelled SV40 cRNA.
