Determination of tryptophan and ten of its metabolites in a single analysis by high-performance liquid chromatography with multiple detection

Abstract

Tryptophan is an essential amino acid whose metabolism involves several pathways. Defects, either inherited or functional, in its transport mechanism or catabolism are related to a large variety of clinical abnormalities. This paper presents a relatively simple and rapid method to determine quantitatively tryptophan and several of its metabolites in biological fluids using reversed-phase high-performance liquid chromatography and multiple detection.

    KYNA has also been determined by UV detection [22,23], or spectrophotometry after derivatization with diazotized sulphanilic acid [24], and less frequently using NMR [25] or tandem mass spectrometry [26,27]. Generally a combination of UV and fluorescent detectors is preferred, although frequently the mobile phases proposed are not compatible with mass spectrometry detectors [28]. Given the lack of studies reporting simultaneous determination by LC of all four compounds in honey, and that the most works are performed on biological matrices [15–28], we sought to develop a solid phase extraction-liquid chromatography with diode array and fluorescence detection (SPE-LC-DAD-FLD) method to simultaneously analysis of TRP, KYNA, KYN and XA in honey samples of distinct botanical origins.
