In inflammatory and demyelinating CNS disorders, Jun et al. (1994) reported that glucocorticoids can modulate the production of NO by microglial cells via the inhibition of protein kinase C (PKC) pathway. Although cortisol may be toxic to neurons, it can also protect neurons by blocking the production of cytotoxic molecules, NO and TNF-α, by microglial cells (Drew and Chavis, 2000).

Recent reports have shown that murine microglial cells can also synthesize NO in response to interferon-γ (IFN-γ) plus LPS or phorbol ester [6,7]. Activity of LPS arises, in part, from the ability of LPS to induce tumor necrosis factor-α (TNF-α) and other cytokines which as a second signal in the activation of macrophage NOS [8].
