Abstract

Simple procedures are described for the preparation of a relatively neutral and of a basic calcium phosphate for column chromatography of proteins.

Bone-bound enzymes for food industry application

The immobilisation of β-galactosidase and amyloglucosidase (AMG) by means of physical adsorption on to bone powder is described. The influence of the enzyme load, applied to the support on immobilisation, yield and efficiency, has been determined. The immobilisation process improved the thermal stability of β-galactosidase (at 37°C), while immobilised AMG displayed a stability (at 55°C) similar to that of the soluble enzyme. The apparent K_M (K_app) values have been determined for both derivatives, for the β-galactosidase derivative K_M was 3 mM using the artificial substrate ONPG and, for the AMG derivative, it was 1 mg/ml using soluble starch as substrate. The extent of lactose hydrolysis achieved, batchwise, with β-galactosidase derivatives acting on lactose buffered solutions, whey, whey permeates and skimmed milk, was in the range of 90% and decreased to 50% after 3 reuses. The conversion of 30% (w/v) liquefied cassava starch achieved with the AMG derivative was 98% until the 11th reuse, and over 95% until the 20th reuse.
Polypeptide γ-subunits of R-phycoerythrin

R-Phycoerythrin was purified from two species of red algae: Callithamnion corymbosum from the Black Sea and Antithamnion sparsum from the Sea of Japan. Three polypeptide γ-subunits differing slightly in molecular weight (31.6, 30.8 and 29.0 kDa) in a 1:5:2 stoichiometry were found in R-phycoerythrin from C. corymbosum by reverse-phase chromatography on a fast performance liquid chromatography (FPLC) system. The ratio of the total number of γ₁-, γ₂- and γ₃-subunits to the total number of α- and β-subunits of R-phycoerythrin was 1:12 and suggested the well-known (αβ)₆γ molecular structure. The chromophore contents of the γ-subunits were identical: three phycourobilins and two phycoerythrobilins. R-Phycoerythrin from A. sparsum also contained three γ-subunits but with different chromophore compositions: one carried three phycourobilins and two phycoerythrobilins while the other carried four phycourobilins and one phycoerythrobilin. The chromophore composition of the third γ-subunit remained unclarified.
Expression of a cytochrome c<inf>2</inf> isozyme restores photosynthetic growth of Rhodobacter sphaeroides mutants lacking the wild-type cytochrome c<inf>2</inf> gene

Deletion of the cytochrome c₂ gene in the purple bacterium Rhodobacter sphaeroides renders it incapable of phototrophic growth (strain cycA65). However, suppressor mutants which restore the ability to grow phototrophically are obtained at relatively high frequency (1–10 in 10⁷). We examined two such suppressors (strains cycA65R5 and cycA65R7) and found the expected complement of electron transfer proteins minus cytochrome c₂: SHP, c′, c_551.5, and c₅₅₄. Instead of cytochrome c₂ which elutes from DEAE-cellulose between SHP and cytochrome c′, at about 50 mm ionic strength in wild-type extracts, we found a new high redox potential cytochrome c in the mutants which elutes with cytochrome c_551.5 at about 150 mm ionic strength. The new cytochrome is more acidic than cytochrome c₂, but is about the same size or slightly smaller (13,500 Da). The redox potential of the new cytochrome from strain cycA65R7 (294 mV) is about 70 mV lower than that of cytochrome c₂. The 280 nm absorbance of the new cytochrome is smaller than that of cytochrome c₂, which suggests that there is less tryptophan (the latter has two residues). In vitro kinetics of reduction by lumiflavin and FMN semiquinones show that the reactivity of the new cytochrome is similar to that of cytochrome c₂, and that there is a relatively large positive charge (+2.6) at the site of reduction, despite the overall negative charge of the protein. This behavior is characteristic of cytochromes c₂ and unlike the majority of bacterial cytochromes examined. Fourteen out of twenty-four of the N-terminal amino acids of the new cytochrome are identical to the sequence of cytochrome c₂. The N-termini of the cycA65R5 and cycA65R7 cytochromes were the same. The kinetics and sequence data indicate that the new protein may be a cytochrome c₂ isozyme, which is not detectable in wild-type cells under photosynthetic growth conditions. We propose the name iso-2 cytochrome c₂ for the new cytochrome produced in the suppressor strains.

This chapter focuses on Phycobiliproteins. In intact cyanobacterial cells and red algal chloroplasts, the phycobiliproteins are components of a complex assemblage, the phycobilisome. The observation that upon cell breakage the products of phycobilisome dissociation are released in water-soluble form gave rise to the unfortunate impression that the phycobiliproteins represented a fairly simple family of well-behaved macromolecules. Appraisal of the true complexity of the situation required an understanding of the molecular structure of phycobilisomes. During purification procedures, certain phycobilisome polypeptide components are lost through retention on chromatographic supports, or, in some cases, destroyed by proteolysis. Some phycobiliproteins, such as allophycocyanin B, are products of subunit scrambling during purification procedures, while others, such as the β^18.3 polypeptide of the phycobilisome core, which is present in only two copies per phycobilisome, may copurify undetected with one of the major phycobiliproteins such as allophycocyanin. A polypeptide pattern obtained by SDS-polyacrylamide gel electrophoresis is an absolutely essential element in the characterization of a purified phycobiliprotein preparation. A sufficient amount of protein should be applied to the gel to ensure that components present in small amounts are detected.

Purification and characterization of polypeptide chain elongation factor 1 from plants

This chapter discusses the procedures for the purification and characterization of elongation factor (EF) EF-l_H, EF-1_L (EF-Iα Guanosine diphosphate GDP), and each EF-1_H subunit from rice embryo, wheat embryo, and cauliflower. The high- and low-molecular-weight forms are designated EF-1_H and EF-1_L, respectively, and the former is thought to be an aggregated form of the latter. EF-l_H from wheat embryo is composed of four subunits, EF-1a, EF-lb, EF-lb', and EF-lc. The process of polypeptide chain elongation in eukaryotes requires two complementary factors, EF-1 and EF-2. EF-1 catalyzes the binding of aminoacyl-transfer RNA (tRNA) to the aminoacyl site on the ribosomes, whereas EF-2 catalyzes the translocation of the newly formed peptidyl-tRNA from the aminoacyl site to the peptidyl site. Cyclic nucleotide-dependent phosphorylation of EF-1_H plays an important role in translational control at the elongation step.
