Age-related changes in the regulation of cholesterol metabolism in rats

Abstract

Activities of the hepatic cholesterol synthetic system including initial steps of the pathway and cholesterol 7α-hydroxylase were all lower in adult (8 to 9-month-old) rats than in young (5 week-old) rats. The extent of diurnal fluctuation of 3-hydroxy-3-methylglutaryl coenzyme A reductase was, however, apparently greater in adult animals. When the cholesterol-enriched diet was fed to rats for 1 day, the extent of the depression of the cholesterogenic enzymes was dependent on age of animals. The enzyme activities rapidly increased on refeeding a cholesterol-free diet after the cholesterol challenge. In young rats the activity of cholesterol 7α-hydroxylase exhibited a pattern inverse to that of HMG-CoA reductase whereas in adult rats it increased continuously during the entire experimental period. Cholesterol and triglyceride accumulated in the liver of adult animals, and their response to dietary cholesterol also depended on the age of the animals. The results indicate a specific modification of the cholesterol homeostatic mechanism with age.

      In contrast, changes in the cholesterol content in thigh meat were subtle (P > 0.05; Figure 1). The reduction of cholesterol with advancing age has been reported by Choi et al. (1987) who found an impairment in enzymes relevant for cholesterol synthesis in adult rats, including acetoacetyl-CoA synthetase, acetoacetyl-CoA thiolase, 3-hydroxyl-3-methylglutryl-coenzyme A (HMG-CoA) reductase, HMG-CoA synthase, mevalonate kinase, and cholesterol 7α-hydroxylase. Moreover, thigh meat showed higher cholesterol content than breast meat in both breeds.

(1987) have found a significant decrease in the enzymatic activity of cholesterol 7α-hydroxylase in rats 5–32 weeks of age. In the study by Wang (2002), gallstone-susceptible C57L mice and resistant AKR mice of both genders split into young adult, older adult, and aged groups (8, 36, and 50 weeks of age, respectively) were each fed a lithogenic diet for 8 weeks.
