Heterocyclic β-Enamino Esters, 13. Halogenation and Rearrangement of Furo[2,3-d]pyrimidinesThe 2,4-dioxo-1,2,3,4,5,6-hexahydrofuro[2,3-d]pyrimidines 3a - e react with phosphoryl chloride not - as expected- to yield the 2-chloro derivatives 4, but instead to form under chlorinating ring cleavage the 6-chloro-5-(2-chloroalkyl)-2,4-dioxo-1,2,3,4-tetrahydropyrimidines 5a - e. Treatment of 5a - d with triethylamine affords in an intramolecular alkylation reaction the 4-chloro-2-oxo-1,2,5,6-tetrahydrofuro[2,3-d]pyrimidines 6a - d, the 4-chloro atom of which can be replaced in a nucleophilic reaction with benzylamine and piperidine to give 7a - d. On the contrary reaction of methylamine with 6a leads to aminolysis yielding 4-chloro-5-(2-hydroxypropyl)-6-methylamino-2-oxo-1-phenyl-1,2-dihydropyrimidine (8), which is obtained likewise from 5a and methylamine. U.v., i.r., and n.m.r. data are described.

Mit Phosphorylchlorid reagieren die 2,4-Dioxo-1,2,3,4,5,6-hexahydrofuro[2,3-d] pyrimidine 3a - e nicht - wie erwartet - zu den 2-Chlor-Derivaten 4, sondern es bilden sich unter chlorierender Ringspaltung die 6-Chlor-5-(2-chloralkyl)-2,4-dioxo-1,2,3,4-tetrahydropyrimidine 5a - e. Bei Einwirkung von Triäthylamin auf 5a - d entstehen durch intramolekulare Alkylierung die 4-Chlor-2-oxo-1,2,5,6-tetrahydrofuro [2.3-d]pyrimidine 6a - d, deren 4-Chloratom mit Benzylamin und Piperidin unter Bildung von 7a - d nucleophil substituierbar ist. Einwirkung von Methylamin auf 6a führt dagegen unter Aminolyse zum 4-Chlor-5-(2-hydroxypropyl)-6-methylamino-2-oxo-l-phenyl-1,2-dihydropyrimidin (8), das ebenfalls aus 5a und Methylamin gewonnen wird. UV-, IR- und NMR-Daten werden beschrieben.
