Ring Transformations in the Isatine Series, IV. SpirocyclizationsThe 4-hydroxy-1,2,3,4-tetrahydro-4-quinazolinecarboxanilides and -N-benzoylcarbohydrazides 6i-I, prepared from the 1-isatinecarboxamides 1b,c by aminolysis, are converted into the spiro-imidazoles 7, the spirooxazole 10, and the spiroindoles 5 by treatment with isocyanates, acetic anhydride, or by heating. The reaction of the (2-acetamidophenyl)glyoxylanilides 2b,c with isocyanates involves competitive ring closure to 5 or to the spiro[benzoxazine-imidazoles] 4. By displacement of the hydroxy group by malononitrile, the reactivity of 6 toward nucleophiles is increased: the esters 6n,o undergo cyclization with o-phenylenediamine to form the spiro[quinazoline-quinoxalines] 11, whereas the anilides or hydrazides 6i-I are readily converted into the spiro[quinazoline-pyrroles] 9.

Durch Aminolyse der 1-Isatincarboxamide 1b,c werden die 4-Hydroxy-1,2,3,4-tetrahydro-4-chinolincarboxanilide und -N-benzoylcarbohydrazide 6i-I hergestellt und mit Isocyanaten, Acetanhydrid bzw. thermisch in die Spiroimidazole 7, das Spirooxazol 10 bzw. die Spiroindole 5 übergeführt. Bei der Reaktion der (2-Acetamidophenyl)glyoxylanilide 2b,c mit Isocyanaten konkurriert der Ringschluß zu 5 mit der Bildung der Spiro[benzoxazin-imidazole] 4. Durch Austausch der Hydroxygruppe gegen Malononitril wird die Reaktivität von 6 gegen Nucleophile gesteigert: die Ester 6n,o cyclisieren mit o-Phenylendiamin zu Spiro[chinazolin-chinoxalinen] 11; die Anilide bzw. Hydrazide 6i-I gehen spontan in die Spiro[chinazolin-pyrrole] 9 über.
