Syntheses of Some β-Desosamine GlycosidesStarting from 3-azido-3-deoxy-1,2;5,6-di-O-isopropylidene-α-D-glucofuranose (1) the 4,6-dichloro-3,4,6-trideoxygalacto derivative 3 has been prepared via 3-azidoglucose 2 by reaction with sulfuryl chloride in pyridine. With titanium tetrabromide the α-pyranosyl bromide 4 is obtained which with different alcohols gave the β-glycosides 5-8 under the conditions of Koenigs-Knorr reaction. Reaction of 5 and 6 with tributyltin hydride yields the 3-acetamido-4,6-dideoxy compounds 15 and 16 from which the amines 11 and 12 have been obtained by treatment with base. Reductive alkylation gave the β-glycosides 13 and 14 of D-desosamine. Without treatment by alkali the same glycosides can be obtained if the acetate group at C-2 in 5 und 6 is removed before tin hydride reduction and reductive alkylation.

Ausgehend von 3-Azido-3-desoxy-1,2;5,6-di-O-isopropyliden-α-D-glucofuranose (1) kann auf dem Wege über 3-Azido-3-desoxy-D-glucose (2) das 4,6-Dichlor-3,4,6-tridesoxygalactoderivat 3 dargestellt werden. 3 reagiert mit Titantetrabromid zum α-Pyranosylbromid 4, das unter den Bedingungen der Koenigs-Knorr-Reaktion mit den entsprechenden Alkoholen die β-Glycoside 5-8 ergibt. Die Umsetzung von 5 und 6 mit Tri-n-butylzinnhydrid führt zu den 3-Acetamido-4,6-didesoxyverbindungen 15 und 16, aus denen durch Basenbehandlung die freien Amine 11 und 12 erhalten werden. Die reduktive Alkylierung führt zu den Desosaminglycosiden 13 und 14. Ohne Alkalibehandlung können die gleichen Glycoside auch erhalten werden, wenn vor der Zinnhydrid-Reduktion und der reduktiven Alkylierung die Acetatgruppe am C-2 von 5 und 6 abgespalten wird.
